Abstract Purpose The relationship between retinal morphology, as assessed by optical coherence tomography (OCT), and function in microperimetry (MP) has not been well studied, despite its increasing importance an essential functional endpoint for clinical trials emerging therapies diseases. Normative databases of healthy ageing eyes are largely missing from literature. Methods Healthy subjects above 50 years were examined using two MP devices, MP‐3 (NIDEK) MAIA (iCare). An identical grid, encompassing 45 stimuli was used sensitivity (RS) assessment. Deep‐learning‐based algorithms performed automated segmentation ellipsoid zone (EZ), outer nuclear layer (ONL), ganglion cell (GCL) nerve fibre (RNFL) OCT volumes (Spectralis, Heidelberg). Pointwise co‐registration each stimulus corresponding location on via registration algorithm. Effect age, eccentricity thickness RS mixed effect models. Results Three thousand six hundred twenty patients included. Mean patient age 68.9 ± 10.9 years. the first exam 28.65 2.49 dB 25.5 2.81 MAIA, respectively. Increased EZ thickness, ONL GCL significantly correlated with increased (all p < 0.001). Univariate models showed lower advanced higher (both 0.05). Conclusion This work provides reference values age‐related ONL‐thicknesses without impairment visual evidence decrease age. information is crucial interpretation future disease.

Geographic atrophy (GA) secondary to age-related macular degeneration is a chronic degenerative disease involving the retinal pigment epithelium, photoreceptors, and choriocapillaris leading irreversible loss of visual function. Identification imaging markers associated with GA development progression has progressed over past decades, moving from two-dimensional three-dimensional imaging, as well image interpretation using artificial intelligence. However, there an open discussion about "must-haves" for detection follow-up complementary imaging. This practical approach provides overview advantages key modalities their applicability in clinical experimental settings.

Inherited retinal degeneration (IRD) disease and age-related macular (AMD) are leading causes of irreversible vision loss blindness. Although significant progress has advanced the field in past 5 years, challenges remain. The current article reviews accomplishments research advances that have fueled development treatments for patients with IRD AMD, including first approved gene-augmentation treatment RPE65-related complement inhibition therapies to slow progression geographic atrophy (GA) AMD. outlines opportunities address gaps unmet needs should lead additional toward IRDs non-neovascular AMD future.
