Accumulation of amyloid-beta (Aβ) plaques leading to oxidative stress, mitochondrial damage, and cell death is one the most accepted pathological hallmarks Alzheimer's disease (AD). Pandanus amaryllifolius, commonly recognized as fragrant screw pine due its characteristic smell, widely distributed in Southeast Asia consumed a food flavor. In search for potential anti-AD agents from terrestrial sources, P. amaryllifolius was explored vitro anti-amyloidogenic neuroprotective effects. Thioflavin T (ThT) assay high-throughput screening multimer detection system (MDS-HTS) were used evaluate extracts' inhibit Aβ aggregations oligomerizations, respectively. The crude alcoholic extract (CAE, 50 μg/mL) base (CBE, obstructed aggregation. Interestingly, results revealed that only CBE inhibited nucleation at 100 μg/mL. Both CAE also restored viability, reduced level reactive oxygen species, reversed dysfunctions 10 20 μg/mL concentrations Aβ-insulted SY-SY5Y cells. addition, unprecedented isolation nicotinamide remarkable discovery bioactive constituents against AD. Hence, our provided new insights into promising extracts AD further exploration other prospective constituents.

Abstract: Alzheimer’s disease (AD) is a progressive, multifactorial, chronic, neurodegenerative with high prevalence and limited therapeutic options, making it global health crisis. Being the most common cause of dementia, AD erodes cognitive, functional, social abilities individual causes escalating medical psychosocial needs. As yet, this disorder has no cure current treatment options are palliative in nature. There an urgent need for novel therapy to address pressing challenge. Digital therapeutics (Dtx) one such that gaining popularity globally. Dtx provides evidence based interventions driven by internet software, employing tools as mobile devices, computers, videogames, apps, sensors, virtual reality aiding prevention, management, ailments like neurological abnormalities chronic diseases. acts supportive tool optimization patient care, individualized improved outcomes. uses visual, sound other non-invasive approaches instance-consistent therapy, reminiscence computerised cognitive training, semantic phonological assistance wearables computer-assisted rehabilitation environment find applications Alzheimer's improving memory, cognition, functional managing motor symptom. A few Dtx-based employed include "Memory Matters", "AlzSense", "Alzheimer Assistant", "smart robotic dog", "Immersive (iVR)" gamma stimulation. The purpose review summarize trends digital explore benefits, challenges, impediments using adjunctive management AD.

Ailanthus altissima is an indigenous plant known for various remedial properties. The present study aimed to evaluate the neuroprotective potential of methanolic extract (AA) bark as current scientific trend searching neurodegenerative diseases, worldwide. In in-vitro experiments, AA was analyzed phenols, flavonoids, antioxidative and cholinesterase inhibitory properties with subsequent detailed characterization secondary metabolites. in-vivo neurological effects were evaluated in rats through behavioral assessment anxiety memory after chronic administration (28 days) 50-200 mg/kg AA. At end behavior studies, isolated brains biochemically tested determine antioxidant enzyme activity.AA found rich phenols/flavonoids active radical scavenging presence 13 metabolites UHPLC-MS analysis. yielded anxiolytic dose-dependently open field, light/dark elevated-plus maze tests animals significantly (P < 0.05 vs control group) preferred arena, illuminated zone exposed arms maze.
