Background: Cholesterol metabolism is important in colorectal cancer (CRC) pathogenesis, influencing tumorigenesis and therapeutic responses. Despite extensive research, fragmented insights a lack of systematic analysis hinder the identification global trends, collaboration networks, emerging hotspots this rapidly evolving field. Methods: A bibliometric 1532 publications (2003–2024) from Web Science Core Collection was conducted using VOSviewer, CiteSpace, R software. Metrics included publication country/institution contributions, author journal influence, keyword co‐occurrence, citation bursts. Results: Global exhibited exponential growth, peaking at 114 2024. The United States (347 articles, 17,613 citations) China (316 articles) dominated research output, yet institutional remained limited. Key journals PLOS One Cancer Research . Keyword evolution shifted “physical activity” “carcinogenesis” to “cholesterol metabolism,” “gut microbiota,” “nanomedicine.” Statins showed preventive potential but raised concerns with prolonged use. Mechanistic highlighted nanoparticle‐driven therapies as promising strategies enhance chemosensitivity reverse immunosuppression. Conclusion: This inaugural comprehensive delineates current landscape cholesterol CRC, particular emphasis on elucidating metabolic reprogramming mechanisms fostering cross‐disciplinary convergence. Future priorities include validating statins’ efficacy via randomized trials, leveraging multiomics for personalized therapies, address geographic disparities translational challenges.

The complex molecular mechanisms involving genetic and epigenetic modifications contribute to colorectal cancer (CRC), which remains a significant threat world health. This review elucidates the role of proteomics genomics in progression, diagnosis, treatment cancers. All potential key pathways involved CRC, including WNT, MAPK, PI3K, TGF-β pathways, are reviewed with systematic analysis, concluding their involvement tumorigenesis therapeutic resistance. Emerging next-generation sequencing technologies revealed critical mutations that relevant CRC development.
