Th1/Th2 balances may play a vital role in the processes of inflammation and fibrosis. The paradigm can be evaluated by representing IFN-γ for Th1 IL-4 Th2. OM-85 BV encouraged preferential development Th1-type immunity characterized amplified decreased production. This study aimed to evaluate inhibitory effect OM85 on bleomycin (BLM)-induced pulmonary fibrosis C57 its possible mechanisms. In vitro experiments demonstrated that exhibited no significant toxicity HELF cells. inhibited TGF-β1-induced protein expression Notch1 Hes1 reduced fibrosis-related marker profiles, such as collagen I, III, fibronectin, P21, α-SMA, following TGF-β1 treatment these Immunofluorescence also revealed α-SMA induced vivo experiments, model was established administering three intratracheal doses BLM (1 mg/kg). BLM-OM85 group exposed an aerosol containing 10.5 mg dissolved 10 mL sterile PBS days 42, 44, 46, 49, 51, 53. BLM-induced fibrosis, leading increased levels lung hydroxyproline, total cell count, macrophages, neutrophils, lymphocytes, well tissue, along with fibrosis-associated proteins α-SMA. Additionally, response suppressed, evidenced bronchoalveolar lavage fluid (BALF), while Th2 amplified, marked BALF. Moreover, morphological assessments showed caused Ashcroft scores, relative content, expanded damaged area, optical density (OD) I. administration significantly mitigated effects. These findings suggest holds therapeutic potential female mice, partly due inhibition modulation IFN-γ/IL-4 ratio.
