Exosomes are extracellular vesicles with diameters of about 100 nm that naturally secreted by cells into body fluids. They derived from endosomes and wrapped in lipid membranes. involved intracellular metabolism intercellular communication. contain nucleic acids, proteins, lipids, metabolites the cell microenvironment cytoplasm. The contents exosomes can reflect their cells' origin allow observation tissue changes states under disease conditions. Naturally have specific biomolecules act as "fingerprint" parent cells, changed pathological conditions be used biomarkers for diagnosis. low immunogenicity, small size, cross blood-brain barrier. These characteristics make unique engineering carriers. incorporate therapeutic drugs achieve targeted drug delivery. carriers therapy still infancy, but exosome provides a new perspective cell-free therapy. Parkinson’s disease (PD) is the second most common neurodegenerative worldwide. Given its prevalence, reliable biomarkers for early diagnosis are required. Exosomal proteins within extracellular nanovesicles promising candidates diagnostic, screening, prognostic, and monitoring purposes in neurological diseases such as PD. This review aims to evaluate potential of vesicle or miRNAs A comprehensive literature search until January 2024 was conducted across multiple databases, including PubMed, EMBASE, Web Science, Cochrane Library, identify relevant studies reporting exosome blood samples from PD patients. Out 417 articles screened, 47 were selected analysis. Among exosomal protein biomarkers, α-synuclein, tau, Amyloid β 1-42, C-X-C motif chemokine ligand 12 (CXCL12) identified significant markers Concerning miRNA miRNA-24, miR-23b-3p, miR-195-3p, miR-29c, mir-331-5p studies. α-synuclein exhibited increased levels patients compared control groups twenty-one studies, while a decrease observed three Our meta-analysis revealed difference total between healthy controls (standardized mean [SMD] = 1.369, 95% confidence interval [CI] 0.893 1.846, p < 0.001), although these results limited by data availability. Furthermore, significantly differ (SMD 1.471, CI 0.941 2.002, 0.001). In conclusion, certain could serve diagnosis, prognosis prediction, assessment progression

Abstract Background The pathogenesis of Parkinson's disease (PD) has not been fully elucidated, and there are no effective disease-modifying drugs for the treatment PD. Mesenchymal stem cells have used to treat several diseases, but readily available. Methods Here, we phenotypically uniform trophoblast stage-derived mesenchymal (T-MSCs) from embryonic cells, which capable stable production, their exosomes (T-MSCs-Exo) explore molecular mechanisms involved in dopaminergic (DA) neuron protection PD models using experimental assays (e.g., western blotting, immunofluorescence immunohistochemistry staining). Results We assessed levels DA injury oxidative stress MPTP-induced mice MPP + -induced MN9D after treating them with T-MSCs or T-MSCs-Exo. Furthermore, T-MSCs-Exo miRNA sequencing analysis revealed that miR-100-5p-enriched directly targeted 3′ UTR NOX4 , could protect against loss neurons, maintain nigro-striatal system function, ameliorate motor deficits, reduce via Nox4-ROS-Nrf2 axis models. Conclusions study suggests may be a promising biological agent Graphical Schematic summary mechanism underlying neuroprotective actions Exo inhibit expression level target gene by delivering miR-100-5p, thereby reducing ROS production alleviating axis, thus improving damage

Abstract Circular RNAs (circRNAs) are a class of covalently closed single-stranded that expressed during the development specific cells and tissues. CircRNAs play crucial roles in physiological pathological processes by sponging microRNAs, modulating gene transcription, controlling activity certain RNA-binding proteins, producing functional peptides. A key focus research at present is functionality circRNAs nervous system several advances have emerged over last 2 years. However, precise role has yet to be comprehensively reviewed. In this review, we first summarize recently described brain development, maturity, aging. Then, on involvement various diseases central system, such as cancer, chronic neurodegenerative diseases, acute injuries neuropathic pain. better understanding will help us develop potential diagnostic, prognostic, therapeutic strategies treat system.

Accumulating evidence suggests that circRNAs play crucial roles in human diseases. CircRNA-disease association prediction is extremely helpful understanding pathogenesis, diagnosis, and prevention, as well identifying relevant biomarkers. During the past few years, a large number of deep learning (DL) based methods have been proposed for predicting circRNA-disease achieved impressive performance. However, there are two main drawbacks to these methods. The first underutilize biometric information data. Second, features extracted by not outstanding represent characteristics between In this study, we developed novel model, named iCircDA-NEAE, predict associations. particular, use disease semantic similarity, Gaussian interaction profile kernel, circRNA expression Jaccard similarity simultaneously time, extract hidden on accelerated attribute network embedding (AANE) dynamic convolutional autoencoder (DCAE). Experimental results circR2Disease dataset show iCircDA-NEAE outperforms other competing significantly. Besides, 16 top 20 pairs with highest scores were validated literature. Furthermore, observe can effectively new potential

Abstract Background Parkinson’s disease (PD) affects 1% of people over 60, and long-term levodopa treatment can cause side effects. Early diagnosis is great significance in slowing down the pathological process PD. Multiple pieces evidence showed that non-coding RNAs (ncRNAs) could participate progression PD pathology. Pyroptosis known to be regulated by ncRNAs as a key feature Therefore, evaluating pyroptosis-related proteins serum worthy biomarkers for early Methods NcRNAs pyroptosis/inflammation mRNA levels were measured with reverse transcriptase quantitative polymerase chain reaction (RT-qPCR). Luciferase assays performed confirm GSDME target miR-675-5p HMGB1 miR-1247-5p. In healthy controls ( n = 106) patients 104), RT-qPCR was utilized assess miR-675-5p, miR-1247-5p, two related (circSLC8A1and lncH19) levels. The enzyme-linked immunosorbent assay 54) 70). Overexpressed or downregulated regulate pyroptosis inflammation Using random forest algorithm, we constructed classifier based on neuron-pyroptosis pathology (four six proteins) having better predictive power than single (AUC 92%). Additionally, verified performance early-stage ≥ 88%). Analysis of transcription factors and chromatin modifications at the genome-wide level provides insights into gene regulatory processes, such as transcription, cell differentiation cellular response. Chromatin immunoprecipitation is most popular powerful approach for mapping chromatin, other enzyme-tethering techniques have recently become available living cells. Among these, Cleavage Under Targets Tagmentation (CUT&Tag) a relatively novel profiling method that has rapidly gained popularity in field epigenetics since 2019. It also been widely adapted to map TFs different species, illustrating association these epitopes with various physiological pathological processes. Scalable single-cell CUT&Tag can be combined distinct platforms distinguish identity, epigenetic features even spatial profiling. In addition, developed strategy joint epigenome, transcriptome or proteome on same sample. this review, we will mainly consolidate applications its derivatives platforms, give detailed explanation pros cons technique well potential development trends future.
