Size exclusion chromatography (SEC) is an effective analytical technique employed for the purification of biomolecules. In size (SEC), biomolecules are sorted according to their size. Objectives: To investigate pectinases from a microbiological source by (SEC). evaluate amount pectinase and total proteins in collected fractions through measurement qualitative analysis. Methods: Utilizing Sephadex G-25 as stationary phase 0.05 M sodium phosphate buffer with increasing concentrations NaCl mobile phase. Using 3,5-dinitro-salicylic acid (DNS) assay pectin-containing agar plate assay, existence that were taken was verified quantitatively subjectively. Results: The order salt concentration 0.15, 0.5, 0.8, 1 1.6 concentration. At 0.15 0.5 concentrations, desired strongly combined Sequential Injection Chromatography (SIC) column eluted at last fraction while chloride early compared buffers containing lower chloride. Conclusions: It concluded suitable enzyme SEC 0.8 because these can be separated very short time low cost.
