Fish represent a large part of the taxonomic diversity vertebrates and are high commercial value. However, factors influencing gut microbiota composition freshwater fish over spatial scales remain unclear. Therefore, this study explored microbiome in 24 species from Yellow River, which spans 1500 km across China. The results showed that geographical distance, host phylogeny diet significantly influenced microbial community diversity, whereas sex, body length weight had minimal influence. Geographical distance was primary factor shaping microbiota, dissimilarity structure increased with an increase mainly driven by dispersal limitation. communities were more homogeneous at higher resolutions due to dominant role selection convergence. Phylosymbiosis observed all species, stronger pattern Cypriniformes, harbour host-specific taxa. Host explained little variation although it significant for metrics tested. Abstract Metformin is a glucose‐lowering drug commonly found in municipal wastewater effluents (MWWEs). The present study investigated the chronic effects of metformin early‐life stages fathead minnow ( Pimephales promelas ). Endpoints assessed were growth, survival, and deformities. larval gut microbiome was also examined using 16 S ribosomal RNA gene amplicon sequencing to determine microbial community composition alpha beta diversity. Eggs larvae exposed measured concentrations (mean [standard deviation]) 0.020 (0.017) μg/L (for controls) 3.44 (0.23), 33.6 (1.6), 269 (11) daily static‐renewal setup, with 20 embryos per beaker. low middle exposure represent river MWWE metformin. To detect small changes we used 18 replicate beakers for controls 9 replicates each treatment. Over 21‐d (5 d as posthatch [dph]), did not affect survival or growth fish. Hatch success, time hatch, deformities hatched fry, similar across all treatments.
