Bisphenol A (BPA) exposure is associated with a plethora of neurodevelopmental abnormalities and brain disorders. Previous studies have demonstrated BPA-induced perturbations to critical neural stem cell (NSC) characteristics, such as proliferation differentiation, although the underlying molecular mechanisms remain under debate. The present study evaluated effects repeated-dose environmentally relevant BPA concentrations during in vitro 3D induction human induced pluripotent cells (hiPSCs), emulating chronic scenario. Firstly, we that our model suitable for NSC differentiation early stages embryonic development. Our morphological image analysis showed at 0.01, 0.1 1 µM decreased average spheroid size by day 21 (D21) induction, while no effect on viability was detected. No alteration rate observed based expression key lineage neuroectodermal transcripts. Quantitative proteomics D21 revealed several differentially abundant proteins across all BPA-treated groups important functions maintenance (e.g., FABP7, GPC4, GAP43, Wnt-8B, TPPP3). Additionally, network alterations glycolytic pathway, potentially implicating changes signalling impairments, well pathophysiology disorders including intellectual disability, autism spectrum disorders, amyotrophic lateral sclerosis (ALS). This enhances current understanding BPA-related aberrations mostly acute, often high dose exposures rodent vivo models GWAS data novel cell-based real-life scenario prolonged low-level exposures, offering further mechanistic insights into ramifications developing consequently, later life neurological

Indoor dust is a key contributor to the global human exposome in urban areas since population develops most of its activities private and public buildings. To gain insight into health risks associated with this chronic exposure, it necessary characterize chemical composition understand biological impacts using reliable physiological models. The present study investigated effects chemically characterized indoor extracts three-dimensional (3D) lung cancer cell cultures combining phenotypic lipidomic analyses. Apart from assessment viability, reactive oxygen species (ROS) induction, interleukin-8 release, lipidomics was applied capture main lipid changes induced as cellular response extracted compounds. application chemometric tools enabled finding associations between compounds lipidic profiles cells. This contributes better understanding toxicity mechanisms exposure pollutants dust.
