In recent decades, the poultry farming industry has assumed a pivotal role in meeting global demand for affordable animal proteins. Ovalbumin (OVA) is the principal protein constituent of eggs. As an alternative to eggs, cell-cultured OVA can reduce environmental impact global warming and land use. Escherichia coli Nissle 1917 (EcN), a probiotic with specific endogenous cryptic plasmids that stably exist in cells without addition antibiotics, was chosen as host for efficient heterologous expression OVA. yield reached 20 mg·L–1 shake flasks using cassette containing tac promoter (Ptac) upstream OVA-coding sequences on plasmid pMUT2. Subsequently, we improved level by employing dual (PP5 combined Ptac via sigma factor binding site 24) ribosome (RBS) substitution. These enhancements increased production 30 42 mg·L–1, respectively. EcNP-P28 harboring L28 equipped both strong RBS8 3.70 g·L–1 3 L bioreactor. Recombinant natural showed similar biochemical characteristics, including secondary structure, isoelectric point, amino acid composition, thermal stability. This currently highest reported among prokaryotes. We successfully constructed antibiotic-free system EcN.
