Abstract In recent years, the field of ancient DNA (aDNA) has taken a new direction toward studying human population dynamics through sedimentary (sedaDNA), enabling study past ecosystems. However, screening numerous sediment samples from archaeological sites remains time-consuming and costly endeavor, particularly when targeting hominin DNA. Here, we present novel high-throughput method that facilitates fast efficient analysis by applying pooled testing method. Our approach involves combining multiple extracts, allowing users to parallelize laboratory procedures early in sample preparation pipeline while effectively for presence aDNA. Pooled exhibit aDNA signals can then undergo detailed analysis, empty pools are discarded. We have successfully applied our extract pooling various Middle Upper Paleolithic Europe, Asia, Africa. Notably, results reveal an signal discernible even with four negative samples. also demonstrate yield double-stranded libraries increases significantly reducing input, potentially mitigating effects inhibition. By embracing this innovative approach, researchers analyze large numbers preservation, achieving significant cost reductions up 70% hands-on time one-fifth.

Understanding the response of long-term aquatic environmental changes in lakes to ongoing climate change and human activities is key forecasting future lake conditions. In this study, we infer Holocene limnological Emu Co, a proglacial Nianbaoyuze on eastern Tibetan Plateau, from sedimentary ancient DNA (sedaDNA) data, palynomorph, element, lithological, grain-size analyses. We developed transfer function based Siberia Tibet/China surface applied it Co sedaDNA trace conductivity changes. The results show that was high during 12.6−9.7 cal ka BP, often surpassing 1000 μs cm−1, driven by elevated summer solar radiation. freshwater influx glacial meltwater precipitation, however, reduced lake's as warmed humidified. This led decrease abundance taxa characterised conductivity. Freshwater pulses, triggered climatic fluctuations, likely significant variations within overarching downward trend. By 8 cal. recharge conditions stabilised reached lower level ∼70 cm−1. warm humid mid-Holocene (8−5 BP) provided suitable habitat for many submerged taxa. After 5 growth restricted, indicated shift asexual sexual reproduction macrophytes, suboptimal colder drier climate. Since 1 might have increased nutrient levels, with an enhanced richness macrophytes. Our indicate how millennial-scale hydrological are related retreat catchment alpine region which today facing much greater than global average.

Accurate land cover data was fundamental for formulating sound planning and sustainable development strategies. This study focused on the Tibetan Plateau (TP), a globally sensitive ecological area, developed locally tailored annual 30 m resolution dataset from 1990 to 2023 (TPLCD). Leveraging Google Earth Engine (GEE) platform Landsat processing, LandTrendr employed generate robust, high-precision training samples. Subsequently, random forest classification spatiotemporal smoothing strategies were applied precisely map dynamics of TP. Lake Limboto, situated in Gorontalo, Indonesia, confronts severe threats jeopardizing its sustainability. Lake ecosystems on the Tibetan Plateau (TP) are highly susceptible to external input owing their fragile environment. However, less study has been done focus direct influences of anthropogenic activities lake sediments by human activities. In this study, we performed first large-scale investigation use fecal stanol proxies in surface assess impact and herbivorous feces TP lakes. The 75 investigated lakes were divided into three types (I, II, III) according cluster analysis ratios R1′ R2′. Fecal stanols type I originate from a mixture feces; II primarily with minimal feces. contrast, III mainly derived (coprostanol concentration > 100 ng/g), mixed source cholesterol reduction 10–100 situ < 10 ng/g). Among investigated, 9 % had significant contamination, 48 moderate or slight 43 uncontaminated. Domestic animal main moderately slightly contaminated central TP. There contamination 52 did not definitely show presence matter. This important implications for assessment provides reference early change paleo-human activity research.

Global climate change has significantly impacted waterbird communities in the warming and humidifying wetlands of Qinghai–Tibet Plateau. However, further research is needed as these transition to warmer drier states. This study focused on diversity richness wintering birds alpine Yamzho Yumco Lake. By analyzing data collected from 17 sampling sites over seven years, we examined abundance changes waterbirds lake surrounding land determined relationship between structure data. The results have shown that abundance, richness, α-diversity index fluctuated. Wintering were positively correlated with monthly mean temperature higher negatively total precipitation water level.
