Algae supplements are widely recognized for their nutritional benefits and commonly marketed as natural health products. However, concerns regarding contamination with cyanobacterial toxins have been raised. Moreover, there is very little data the potential of algal on Chinese market by these toxins. In this study, we employed a validated solid-phase extraction ultra-high-performance liquid chromatography-tandem mass spectrometry (SPE-UHPLC-MS/MS) method to analyze available in China. Therefore, study optimized conditions twelve microcystins (MCs) simultaneously using response surface methodology (RSM) analyzed via UHPLC-MS/MS. Method validation was conducted four supplement matrices (capsule, liquid, powder, tablet) ensure method's accuracy, sensitivity, reproducibility. Analysis nineteen commercial algae products China revealed presence MCs: microcystin-LF (MC-LF), microcystin-LR (MC-LR), microcystin-LA (MC-LA), microcystin-YR (MC-YR). Furthermore, seven were found contain one or more MCs, two exceeding 1 μg/g MC limit. These findings underscore effectiveness assessing consequently identifying consumers expected be at risk from prolonged exposure recommended daily supplements.

Microcystin-LR (MC-LR) a cyclic toxin produced by cyanobacterial species is known to exert detrimental effects on various organs, including lung. Several investigators demonstrated that MC-LR exerts pulmonary toxicity, but the underlying mechanisms remain unclear. This study aimed investigate whether exposure MC-LR-induced lung inflammation and examine mechanisms. Thirty specific pathogen-free (SPF) male mice were allocated into control treatment groups. Mice intraperitoneally injected with physiological saline or (20 μg/kg) daily for total of 21 days. Our findings indicated MC-LR-produced histopathological changes in tissue, thickening alveolar walls inflammatory infiltration. was found upregulate mRNA expression levels pro-inflammatory cytokines TNFα, IL-6, IL-1β, IL-18. Further, significantly elevated proteins associated NF-κB/NLRP3 pathway p-NF-κB, NLRP3, Caspase-1, ASC. The activation further promoted release cytokine IL-1β cleavage pyroptosis-associated GSDMD protein. These indicate may induce promoting cell pyroptosis via pathway.

The global phenomenon of cyanobacterial bloom pollution is spreading globally due to climate change and eutrophication. It well established that harmful cyanobacteria produce a wide range toxins including microcystin-LR (MC-LR), cyclic heptapeptide toxin known damage various organs. intestinal tract the main site MC-LR absorption one targets susceptible toxicity. Currently, studies on enterotoxic effects predominantly focused colorectum, with limited investigations addressing impact microcystins small intestine. Therefore, aim our study was examine chronic 9-month exposure mice low-dose 120 μg/L in drinking water ileal inflammation potential mechanisms underlying these effects. Our findings showed chronically administered disorganized epithelial cells, lymphocytic infiltration disturbed crypt arrangement were detected. results qPCR Western blot demonstrated that, comparison control, mRNA expression levels pro-inflammatory factors IL-6, IL-17, IL-18, IFN-γ markedly elevated tissue treated MC-LR, associated significant increases protein p-PI3K, p-AKT, p-mTOR. Taken together, evidence indicates induces histopathological involved activation PI3K/AKT/mTOR signaling pathway.

ABSTRACT Abnormal bile acid (BA) metabolism is involved in liver fibrosis. In a previous study, we discovered that the hydrophobic BA glycochenodeoxycholate (GCDCA) induced fibrosis and GW4064, an agonist of farnesoid X receptor (FXR), alleviated caused by GCDCA. However, impacts GCDCA on BAs, gut intestinal barrier, microbiota are unclear, obtaining this information would provide additional into role development present ultra-performance liquid chromatography‒tandem mass spectrometry revealed mice administered gavage had higher levels total primary BAs than those control group, significant reduction was observed + GW4064 group compared with group. Compared greater gut, especially T-alpha-MCA T-beta-MCA, no differences terminal ileum were between groups. Immunohistochemistry indicated administration inhibited FXR FGF15 expression, whereas activated promoted expression. Moreover, immunohistochemistry decreased mucin2, claudin-1, occludin, ZO-1 restored their 16S rDNA sequencing alpha diversity did not significantly differ among three groups, but beta At phylum level, disturbed microbiota, as reductions Desulfobacterota, Bacteroidota, Actinobacteria increased abundances Proteobacteria, Cyanobacteria, Patescibacteria noted improved structure at level. Accurate detection and monitoring of environmental pollutants are paramount importance for disease prevention public health. In recent years, the ever-expanding human activities industrial production have given rise to a sharp increase in complexity variety these pollutants, which pose significant threats well - being. Environmental stem from multiple sources, such as heavy metals, persistent organic inorganic non metallic emerging biological contaminants. Traditional technologies, though valuable their sensitivity accuracy, constrained by complex sample preparation, poor selectivity, absence standardized methods. On other hand, including nanotechnology, molecular methods, biosensors, Surface-Enhanced Raman Spectroscopy (SERS), multi-omics, big data analysis, offer promising solutions rapid sensitive pollutant detection. The establishment networks sharing platforms further enhances real time provides solid support health initiatives. Nonetheless, challenges persist, integration, exposure assessment, development cost-effective portable solutions. Future progress interdisciplinary approaches technology integration will be crucial advancing facilitating comprehensive prevention. This review systematically classifies showcases latest advancements offering critical insights protection.
