Abstract Pine wilt disease (PWD), caused by the pine wood nematode (PWN) Bursaphelenchus xylophilus , results in significant economic and ecological damage to Pinus forests plantations worldwide. massoniana is primary host of PWD southern China, but its response PWN remains largely unstudied. Previously, we observed a P. nursery that contained over 71 commonly used cultivars. Through field phenotyping, identified two groups resistant Resistant cultivars (RC) exhibited very low carrying amounts (PCA) had relatively mortality, tolerant (TC) high PCA mortality. In this study, confirmed via inoculation assays lower mortality rates, 10% 11%, respectively, than other (which rate 83.3%). The RC was significantly cultivars, while TC higher PCA. To explore molecular mechanisms underlying PWN, transcriptome metabolome above were profiled throughput sequencing. Second, inhibiting aspartic acid, arginine proline APS5 (aspartate aminotransferase 5) suppressed. Third, reducing cysteine, GERD (germacrene D synthase) up-regulated, sesquiterpenoid triterpenoid metabolites accumulated resist invasion. 2) most effective accumulation reactive oxygen species (ROS), which enhanced jasmonic (JA) highly induced expression chitinase, improving PWN. 3) tolerance induction phosphatidylcholine, promoted flavonoid anthocyanin LPIN (phosphatidate phosphatase), LOX2S (lipoxygenase), LOX1_5 (linoleate 9S-lipoxygenase), pathogenicity not These illustrate or tolerate are great significance prevention control PWD.
