Introduction Diuron (3-(3,4-dichlorophenyl)-1,1-dimethylurea) is a prominent herbicide in Brazilian agriculture, particularly sugarcane farming. This persists the environment and transformed into potentially toxic metabolites, such as 3,4-dichloroaniline (DCA) 3-(3,4-dichlorophenyl)-1-methylurea (DCPMU), raising concerns that encompass environmental contamination One Health, which unites human health. Zebrafish are an appropriate experimental model to evaluate toxicity: they metabolically genetically resemble humans can be applied ecotoxicology. Methods In this study, we have employed multiparameter evaluation characterize toxicity of environmentally relevant concentrations (0.5–100 μM) Diuron, DCA, or DCPMU Brazil, developing country. early stages zebrafish development, performed fish embryo test according guidelines established OECD Protocol No. 236, extended 144 h post fertilization (hpf). We also investigated oxygen consumption rate embryos at 24 hpf, evaluated acetylcholinesterase (AChE) enzymatic activity larvae 96 used Comet Assay assess DNA fragmentation hpf. subjected adult acute exposure for accordance with protocol no. 203 Acute Toxicity Test Fish. Following exposure, analyzed mutagenicity by using micronucleus test, was complemented quantification nuclear anomalies (NA) erythrocytes further confirmed histopathological investigations. Results discussion Exposure any tested damaged DNA, NA became significant beyond 10 μM Diuron. At μM, DCA altered AChE activity, suggesting neuromuscular toxicity. Between 0.5 5 genotoxic, 5–10 induced pleomorphism. all concentrations, emerged between 1 DCPMU. Adult exposed did not exhibit alterations. Erythrocyte morphology analysis reveal mutagenicity. Although observed non-mutagenic genotoxicity, chromosomal instability concern. The findings study contribute our understanding how its metabolites affect highlights need multiparametric assessment elucidate herbicides their impact endeavor informs regulatory measures safely similar reveals substantial implications nations grappling escalating water due pesticide use.
