Per-and polyfluoroalkyl substances (PFASs) affect thyroid function, impairing neonatal development and growth. This study aims to explore the association between PFASs hormones in umbilical cord blood of neonates delivered spontaneously. A total 119 puerperae who vaginally were included. Twenty-nine quantified plasma using a Waters ACQUITY ultra-performance liquid chromatography (UPLC) system coupled with Quattro Premier XE triple quadrupole mass spectrometer. Five Roche Analytics E170 modular analyzer. Perfluorooctanoic acid (PFOA), 6:2 chlorinated polyfluorinated ether sulfonic (6:2 Cl-PFESA), linear perfluorooctane (L-PFOS) present highest levels median (quartile 1-quartile 3) 3.23 (2.32-4.32), 1.35 (0.84-2.01), 0.94 (0.63-1.41) ng/mL, respectively. The regression analysis revealed that perfluorohexane (L-PFHxS) (β = 0.557, p 0.038) perfluorononanoic (PFNA) 0.613, 0.045) independently positively associated free triiodothyronine (T3), but PFOA exhibited an inverse trend -0.040, 0.002). sum 3,4,5 monohydroperfluorooctane sulfonates (Σ3,4,5 m-PFOS) was negatively T3 -0.349, 0.007). Per- and polyfluoroalkyl substances (PFASs) are among the persistent organic pollutants characterized by their persistence in environment, high mobility, adverse impact not only on ecosystem but also human health. The biggest challenges biomonitoring low concentrations of PFASs biological matrices presence matrix interferents samples. combination liquid chromatography with tandem mass spectrometry (LC-MS/MS) solid-phase extraction (SPE) as a sample preparation technique appears to be most suitable solution for achieving desired selectivity sensitivity PFAS determination. aim this review is describe possible sources PFASs, various matrices, analytical methods determining different using pretreatment techniques complex samples, well health risks associated exposure. studied include PFOA PFOS, which frequently detected such plasma, serum, breast milk. average range from 1.0 2.6 ng.mL−1 1.9 2.4 0.4 3.1 For were 2.0–4.0 ng.mL−1, 3.7–4.6 3.6–4.8 milk, respectively. significant effects exposure long-chain (such PFOS) lipid disorders, hypertension, diabetes mellitus, thyroid infertility, cancer, obesity, autism, neurodevelopmental issues, cardiovascular diseases, kidney liver disorders. Perfluoroalkyl and polyfluoroalkyl substances (PFASs) are emerging contaminants widely found in drinking water food, potentially entering the human body through these sources. To investigate effects, we simulated PFAS exposure doses animal models, ranging from general population to occupational levels, analyzed blood spleen samples. Epidemiological studies linked immunosuppression hematotoxicity, while toxicogenomic analyses validated underlying biological mechanisms. caused immunotoxic including altered parameters lymphocyte edema. Cellular abnormalities such as decreased cytoplasmic density, incomplete rough endoplasmic reticulum, chromatin condensation, nuclear shrinkage, reduced mitochondrial cristae were observed. evidence revealed a dose-response relationship between mixed hematologic indices hematocrit (HCT), hemoglobin (HGB), platelet count (PLT), platelet-to-lymphocyte ratio (PLR), white cell (WBC), monocyte percentage, large (PLCR), with perfluorooctanoic acid (PFOA) playing dominant role. Toxicogenomic analysis identified genes associated platelets, anemia, leukocyte function, inflammation, Th17 differentiation, apoptosis, lipid metabolism, atherosclerosis, JAK-STAT signaling. This study provides novel insights into hematotoxicity immunosuppressive effects of exposure, emphasizing need for substitution, removal, policies addressing exposures protect public health.
