The effects of perfluoroalkyl and polyfluoroalkyl substances (PFAS) on genomic stability remain unclear. Here, a cross-sectional study was conducted to establish the associations PFAS with genotoxic biomarkers. We recruited cohort 453 residents in 2021 Zhejiang, China. Thirty serum were quantified, alongside seven indicators [five rDNA copy numbers (rDNA-CN), mitochondrial DNA (mtDNA-CN), relative telomere length (RTL)] whole blood. Results showed that PFUnDA, perfluorohexanesulfonic acid (PFHxS), perfluorooctanesulfonic (PFOS), 6:2 Cl-PFESA, PFO5DoDA positively correlated rDNA-CN, while PFHpA, PFOA, PFMOAA inverse associations. PFO4DA mtDNA-CN. HFPO-TA, negatively associated RTL, perfluorononanoic acid, PFHxS, PFOS, Cl-PFESA positive Nonlinear exposure-response relationships also observed between biomarkers using restricted cubic spline models. Furthermore, mixtures mtDNA-CN, showing highest contribution by quantile-based g-computation model. In vivo studies further confirmed increased mtDNA-CN male mice dose-dependent manner. This provides novel evidence disrupt stability, varying functional groups fluoroalkyl(ether) chain lengths.

Fluoroether approach has been extensively employed as an alternative to conventional fluorinated surfactants; however, it faces challenges such inadequate performance of short fluoroether chains and environmental contamination from long chains. This study aims address the conflict between compatibility surfactants. The design surfactants is centered around OC chain (CF3(OCF2)n–), utilizing reduced fluorine content achieve superior surface activity. A series surfactant molecules featuring elongated hydrophobic dual-chain structures have synthesized characterized. All good activity at low concentrations. Among them, CF3(OCF2)4CH2O(CH2)3SO2NH(CH2)3N+(CH3)2CH2COO– (OC4F-B), which employs a strategy extending by insertion –CH2O(CH2)3–, shows best performance. can tension 20 mN/m concentration 0.005 wt %, times lower than that required for CF3(OCF2)4CONH(CH2)3N+(CH3)2CH2COO– (OC4-betaine) attain same tension. In contrast, (CF3(OCF2)3CH2O(CH2)3SO2NHCH2CH2)2N+(CH3)CH2COO– (DOC3F-B), both extended carbon strategies, exhibits critical micelle diminished 2 orders magnitude relative OC3-betaine, reaching 0.047 g/L. Merely adjusting length fluorocarbon insufficient reconcile impact introduces innovative incorporating hydrocarbon adopting design, thereby addressing this challenge effectively.

Concentrations of 48 per- and polyfluoroalkyl substances (PFAS) were measured in settled dust samples from 65 homes GenX Exposure Study participants residing near a fluorochemical manufacturing facility North Carolina. Eight PFAS [perfluoro(3,5-dioxahexanoic) acid (aka PFO2HxA), perfluoro-2-(perfluoromethoxy)propanoic (PMPA), perfluoro-2-ethoxypropanoic (PEPA), 6:2 fluorotelomer phosphate diester (6:2 diPAP), 6:2/8:2 (6:2/8:2 perfluoropropanoic (PFPrA), perfluorodecanesulfonic (PFDS), 2-(N-ethyl-perfluorooctanesulfonamido)acetic (N-EtFOSAA)] detected >90% the samples. Dust concentrations six ether acids (PFEAs) produced at (PEPA, PMPA, perfluoro-2-methoxyacetic (PFMOAA), PFO2HxA, hexafluoropropylene oxide dimer (HFPO-DA aka GenX), ethanesulfonic acid, 2-[1-[difluoro(1,2,2,2-tetrafluoroethoxy)methyl]-1,2,2,2- tetrafluoroethoxy]-1,1,2,2-tetrafluoro- (Nafion byproduct 2) significantly negatively associated with home distance facility. Homes closer to had higher summed mass 12 targeted PFEAs (∑12PFEAs) but not ∑48PFAS. Trifluoroacetic (TFA), an ultrashort chain PFAS, three diPAPs (8:2/6:2 diPAP, 8:2 diPAP) major contributors ∑48PFAS, these compounds did show relationships distance.
