Temporal and cross-sectional associations of serum per- and polyfluoroalkyl substances (PFAS) and lipids from 1986 to 2016 − The Tromsø study

Per-
and
polyfluoroalkyl
substances
(PFAS)
have
been
linked
to
effects
on
human
lipid
profiles,
with
several
epidemiological
studies
reporting
associations
between
specific
PFAS
blood
concentrations.
However,
these
inconsistent,
most
focused
cross-sectional
analyses
limited
repeated
measurements.
In
this
study,
we
investigated
serum
concentrations
major
classes
over
a
30-year
period
(1986-2016)
up
five
time
points.
Lipids
analyzed
included
total
cholesterol
(TC),
low-density
lipoprotein
(LDL-C),
high-density
(HDL-C),
triglycerides
(TG).
This
study
145
participants
from
The
Tromsø
Study,
Norway,
who
donated
plasma
samples
three
times
the
period.
Linear
mixed-effects
(LME)
models
assessed
longitudinal
classes,
while
multiple
linear
regression
(MLR)
were
used
for
associations.
LME
demonstrated
positive
perfluorooctanoic
acid
(PFOA),
perfluorononanoic
(PFNA),
perfluorodecanoic
(PFDA),
perfluoroundecanoic
(PFUnDA),
perfluorododecanoic
(PFDoDA),
perfluorotridecanoic
(PFTrDA)
TC.
Additionally,
PFOA,
PFDA,
PFUnDA,
PFDoDA,
PFTrDA
associated
LDL-C,
PFUnDA
summed
perfluorooctane
sulfonate
isomers
(∑PFOS)
HDL-C.
Cross-sectional
corroborated
six
compounds
TC
at
least
times,
but
LDL-C
HDL-C
not
confirmed.
Summed
sulfonamide
(∑PFOSA)
showed
negative
association
longitudinally,
was
confirmed
cross-sectionally.

Abstract
Per‐
and
polyfluoroalkyl
substances
(PFAS)
are
ubiquitous
persistent
organic
pollutants.
The
neurotoxic
cardiometabolic
effects
of
PFAS
well
documented,
leading
to
the
hypothesis
that
exposure
increases
dementia
risk.
However,
empirical
data
on
in
relation
cognitive
impairment
weak,
limited,
inconsistent.
This
report
reviews
literature
provides
a
rationale
overview
VascCog
Longitudinal
Study,
new
study
using
population‐based
Northern
Manhattan
Study
cohort,
prospectively
examine
serum
concentrations
13
lipids,
carotid
atherosclerosis,
impairment,
dementia.
We
hypothesize
deleteriously

cognition

hyperlipidemia
atherosclerosis.
