Polyphenols were extracted from Thuja occidentalis L. bark and characterized in terms of phytochemical composition biological activity. The optimization process investigated the impacts extraction duration, ultrasound amplitude, ethanol concentration on total phenolic content extract. antioxidant capacity was examined using DPPH ABTS assays, anti-enzyme activity against alpha-glucosidase, alpha-amylase, lipase, acetylcholinesterase, tyrosinase determined. experimental model revealed optimal parameters: a hydroethanolic solvent with 44% v:v ethanol, 15-minute time, 40% amplitude. These parameters validated confirmed by reaching target Total Phenolic Content (TPC) predicted MODDE software. resulting lyophilized extract exhibited high polyphenolic (161.84 ± 23.48 mg GAE/g dry extract) demonstrated strong properties. Notably, it showed significant inhibition alpha-glucosidase suggesting potential antidiabetic effects, though its relatively weak. findings provide foundation for further investigations to confirm qualitative quantitative presence specific metabolites elucidate mechanisms underlying bioactive
