Lycopene (LYC) is a carotenoid, has antioxidant properties. This study investigated whether lycopene attenuates aristolochic acids (AAs) -induced chronic kidney disease. In this experiment, was used to intervene C57BL/6 mice with renal injury induced by acid exposure. The histomorphological changes and serological parameters of the were measured in order assess alleviating effect on nephropathy. vitro vivo experiments carried out verify main mechanism action drug targets improving nephropathy (AAN) various experimental methods such as ELISA, immunohistochemistry, immunofluorescence, Western-blot qRT-PCR. results showed that oxidative stress after AAI exposure, resulting inflammatory response tubular epithelial cell apoptosis. Nrf2/HO-1 signaling pathway inhibited induces kidney, which ultimately leads inflammation After LYC intervened body, it activated Nrf2 nuclear translocation its downstream HO-1 NQO1 pathways. ROS production cells, alleviated mitochondrial damage. further modulated TNF-α/NF-κB cascade, thereby reduced accumulation factors interstitium. Moreover, able up-regulate expression Bcl-2, down-regulate Bax inhibit activation cleaved forms Caspase-9 Caspase-3, finally attenuated apoptosis It concluded activate maintain homeostasis These suggested can be relieve AAN.

Bioactive compounds represent a broad class of dietary metabolites derived from fruits and vegetables, such as polyphenols, carotenoids glucosinolates with potential for cancer prevention. Curcumin, resveratrol, quercetin, β-carotene have been the most widely applied bioactive in chemoprevention. Lately, many approaches to encapsulating components nano-delivery systems improved biomolecules' stability targeted delivery. In this review, we critically analyze compounds, including polymeric nanoparticles (NPs), solid lipid (SLN), nanostructured carriers (NLC), liposomes, niosomes, nanoemulsions (NEs) use therapy. Efficacy studies nanoformulations using cell lines vivo models updated human clinical trials are also discussed. Nano-delivery were found improve therapeutic efficacy molecules against various types (e.g., breast, prostate, colorectal lung cancer) mainly due antiproliferation pro-apoptotic effects tumor cells. Furthermore, some promised combination therapy standard chemotherapeutic agents, increased efficiency fewer side effects. These opportunities identified developed ensure more excellent safety novel herbal medicines enabling insights designing

Renal fibrosis is a typical pathological change in chronic kidney disease (CKD). Epithelial-mesenchymal transition (EMT) the predominant stage. Activation of macroautophagy/autophagy plays crucial role process EMT. Lycopene (LYC) highly antioxidant carotenoid with pharmacological effects such as anti-inflammation, anti-apoptosis and mediation autophagy. In this study, we demonstrated specific mechanism LYC activating mitophagy improving renal fibrosis. The enrichment analysis results GO KEGG showed that had high values alleviated aristolochic acid I (AAI)-induced intracellular expression PINK1, TGFB/TGF-β, p-SMAD2, p-SMAD3, PRKN/Parkin, recruited MAP1LC3/LC3-II SQSTM1/p62, decreased mitochondrial membrane potential (MMP), ameliorated mice. When simultaneously intervened NRK52E cells using bafilomycin A1 (Baf-A1), AAI, LYC, MAP1LC3-II SQSTM1 was significantly increased. A similar result seen tissue when treated vitro vivo CQ, inhibitory effect on AAI-activated SMAD2-SMAD3 signaling pathway attenuated. Molecular docking simulation experiments stably bound to AKT active site. After intervention AAI GSK-690693, PRKN, MAP1LC3-II, BECN1, p-SMAD2 p-SMAD3 increased, decreased. However, SC79 inhibited autophagy reversed could inhibit activate reduce

The positive effect lycopene the result its pleiotropic effect. ability to neutralize free radicals via one foundations pro-health effect, including inhibit development many civilization diseases. Therefore, this study focuses on importance in inhibiting diseases such as cardiovascular diseases, within nervous system, diabetes, liver and ulcerative colitis. According research mentioned, supplementation has significant promise for treatment illnesses marked by chronic inflammation oxidative stress. However, majority supporting data lycopene′s health benefits comes from experimental research, whereas evidence clinical studies both scarcer less certain any benefits. Research humans still required establish effectiveness.

Aflatoxin (AFT) contamination poses a significant global public health and safety concern, prompting widespread apprehension. Of the various AFTs, aflatoxin B1 (AFB1) stands out for its pronounced toxicity association with spectrum of chronic ailments, including cardiovascular disease, neurodegenerative disorders, cancer. Lycopene, lipid-soluble natural carotenoid, has emerged as potential mitigator deleterious effects induced by AFB1 exposure, spanning cardiac injury, hepatotoxicity, nephrotoxicity, intestinal damage, reproductive impairment. This protective mechanism operates reducing oxidative stress, inflammation, lipid peroxidation, activating mitochondrial apoptotic pathway, facilitating activation biogenesis, endogenous antioxidant system, nuclear factor erythroid 2-related 2 (Nrf2)/kelch-like ECH-associated protein 1 (KEAP1) peroxisome proliferator-activated receptor-γ coactivator-1 (PGC-1) pathways, well regulating activities cytochrome P450 (CYP450) enzymes. review provides an overview lycopene against exposure-induced underlying molecular mechanisms. Furthermore, it explores profile clinical applications lycopene. The present underscores lycopene’s promising detoxification agent intent to stimulate further research practical utilization in this domain.

Abstract The high incidence of oral cancer combined with excessive treatment cost underscores the need for novel preventive and therapeutic options. value natural agents, including plant secondary metabolites (phytochemicals), in preventing carcinogenesis representing expansive source anticancer drugs have been established. While fragmentary research data are available on antioral effects phytochemicals, a comprehensive critical evaluation potential these agents prevention intervention human malignancies has not conducted according to our knowledge. This study presents complete analysis current preclinical clinical results using phytochemicals. Our in‐depth highlights various such as phenolics, terpenoids, alkaloids, sulfur‐containing compounds, against numerous cells and/or vivo models by antiproliferative, proapoptotic, cell cycle‐regulatory, antiinvasive, antiangiogenic, antimetastatic effects. Bioactive phytochemicals exert their antineoplastic modulating signaling pathways, specifically involving epidermal growth factor receptor, cytokine receptors, toll‐like tumor necrosis receptor consequently alter expression downstream genes proteins. Interestingly, demonstrate encouraging trials, reduction lesion size, growth, pain score, development new lesions. most displayed minimal toxicity, concerns bioavailability may limit application. Future directions include more mechanistic studies, administration formulations, investigation phytocompounds adjuvants conventional treatment, randomized trials.
