The role of the microbiome in immunotherapy has recently garnered substantial attention, with molecular studies and clinical trials providing emerging evidence on pivotal influence microbiota enhancing therapeutic outcomes via immune response modulation. However, impact microbial communities can considerably vary across individuals different immunotherapeutic approaches, posing prominent challenges harnessing their potential. In this comprehensive review, we outline current research applications tumor delve into possible mechanisms through which function is influenced by various body sites, encompassing those gut, extraintestinal barrier, intratumoral environment.Furthermore, discuss effects diverse microbiome-based strategies, including probiotics, prebiotics, fecal transplantation, targeted modulation specific taxa, antibiotic treatments cancer immunotherapy. All these strategies potentially have a profound pave way for personalized approaches predictive biomarkers.

Abstract Background Prostate cancer (PCa) incidence and mortality rates are rising. Our previous research has shown that the combination of icariin (ICA) curcumol (CUR) induced autophagy ferroptosis in PCa cells, altered lipid metabolism. We aimed to further explore effects ICA CUR on gut microbiota, metabolism, immunity PCa. Methods A mouse subcutaneous RM-1 cell tumor model was established. 16 S rRNA sequencing performed detect changes fecal microbiota. SCFAs feces, effect ICA-CUR T-cell immunity, IGFBP2, DNMT1 were examined. Fecal microbiota transplantation (FMT) conducted mechanism ICA-CUR. Si-IGFBP2 si/oe-DNMT1 transfected into DU145 cells treated with investigate development. Results After treatment ICA-CUR, there a decrease volume weight, accompanied by affected DNMT1/IGFBP2/EGFR/STAT3/PD-L1 pathway. increased positive CD3 + CD8 IFN-γ, Ki67 levels IFN-γ IFN-α serum. FMT (with donors from group), weight decreased. promote development expression IGFBP2. In vitro, DNMT1/IGFBP2 promotes migration proliferation. inhibits DNMT1/IGFBP2. Cancer remains a public health concern worldwide, with its incidence increasing worldwide and expected to continue growing during the next decades. The microbiome has emerged as central factor in human disease, demonstrating an intricate relationship between cancer. Although some microbiomes present within local tissues have been shown restrict cancer development, mainly by interacting cells or host immune system, microorganisms are harmful risk factors for development. This review summarizes recent evidence concerning of most common types (i.e., lung, head neck, breast, gastric, colorectal, prostate, cervix cancers), providing general overview future clinical approaches perspectives.

Immune checkpoint inhibitors (ICIs), such as durvalumab, tremelimumab, and atezolizumab, have emerged a significant therapeutic option for the treatment of hepatocellular carcinoma (HCC). In fact, efficacy ICIs single agents or part combination therapies has been demonstrated in practice-changing phase III clinical trials. However, confront several difficulties, including lack predictive biomarkers, primary secondary drug resistance, treatment-related side effects. Herein, we provide an overview current issues future challenges this setting.

To create a diagnostic tool before biopsy for patients with prostate-specific antigen (PSA) levels < 20 ng/ml to minimize prostate biopsy-related discomfort and risks. Data from 655 who underwent transperineal at the First Affiliated Hospital of Wannan Medical College July 2021 January 2023 were collected analyzed. After applying Synthetic Minority Over-sampling TEchnique class balancing on training set, multiple machine learning models constructed by using Least Absolute Shrinkage Selection Operator (LASSO) feature selection identify significant variables. The best-performing model was selected evaluated through tenfold cross-validation ensure interpretability. Finally, performance assessed test set data validation. age, mass ratio (PSAMR), Prostate Imaging–Reporting System, volume as variables construction based LASSO regression. receiver operating characteristic (ROC) results in validation follows: XGBoost: 0.93 (0.88–0.97); logistic: 0.89 (0.83–0.95); LightGBM: 0.87 (0.80–0.93); AdaBoost: 0.90 (0.85–0.96); GNB: 0.88 (0.82–0.95); CNB: 0.79 (0.71–0.87); MLP: 0.78 (0.69–0.86); Support Vector Machine: 0.81 (0.73–0.89). XGBoost best reconstructed data, resulting following ROC scores: 0.995 (0.991–0.999), 0.945 (0.885–0.997 ), 0.920 (0.868–0.972). Kolmogorov–Smirnov curve, calibration curve yielded positive results; decision demonstrates that threshold probabilities ranging 10 95% can benefit this model. Prostate cancer is one of the most common cancers in United States with a high mortality rate. In recent years, traditional opinion about prostate microbiome was challenged. Although there still are some arguments, an escalating number researchers shifting their focus toward within tumor environment. We mined data extracted from metagenome, and it offers broader taxonomic coverage accurate functional profiling. used Kraken2, mapping tool, to mine gut microbiota patients. A two-sample Mendelian Randomization conducted reflect association between cancer. study, we found consistency special intratumor both non-metastatic tumors metastatic tumors. ABSTRACT Cancer‐associated fibroblasts (CAFs) are key stroma cells that play dominant roles in the migration and invasion of several types cancer through secretion inflammatory cytokine IL‐17A. This study aims to identify potential role regulatory mechanism CAFs‐secreted IL‐17A prostate (PC). CAFs normal (NFs) were obtained from fresh PC its adjacent tissues, respectively. LNCaP DU145 co‐cultured with conditioned medium NFs. level was determined by ELISA cell supernatant. CCK‐8, wound healing, Transwell assay, western blot analysis, staining, primary lung metastasis assays employed vivo or vitro explore effect secreted them on proliferation, migration, invasion, epithelial−mesenchymal transition (EMT) PC. stimulated cells. Importantly, exerted their directly secreting IL‐17A, leading a significant increase Mechanically, promoted Smad3/p38 MAPK pathway‐mediated EMT process, contributing invasion. Furthermore, activated pathway thus facilitated tumor growth nude mice. identifies novel signaling which mediate via upregulation MAPK‐mediated an IL‐17A‐dependent manner.
