Cancer is a mysterious disease. Among other alterations, tumor cells, importantly, have metabolic modifications. A well-known modification commonly observed in cancer cells has been termed the Warburg effect. This phenomenon defined as high preference for glucose uptake, and increased lactate production from that glucose, even when oxygen readily available. Some anti-cancer drugs target proposed effect, some dietary regimens can function similarly. However, most suitable strategies treating particular cancers are not yet well understood. The aim of this review was to describe findings regarding impact various targeting evidence suggests combining routine therapies with diet-based may improve outcome cancer. designing individualized must be our ultimate goal.

Abstract Combining metabolic therapy with photothermal (PTT) shows promise for the treatment of triple‐negative breast cancer (TNBC). However, a strategy is still required to effectively overcome adaptation TNBC thermotolerance during thermotherapy, thereby enhancing therapeutic outcomes. In this study, polydopamine‐integrated nanomedicine designed, glucose oxidase (GOx) chemically conjugated its surface and curcumin (Cur)‐loaded calcium phosphate (CaP) shell. The outer CaP layer disintegrates in response acidic environment tumor cells, releasing ions efflux inhibitor Cur, which causes mitochondrial functional disruption by inducing overload. Concurrently, GOx consumes intracellular inhibit glycolysis. dual intervention metabolism glycolysis serves counteract adaptations TNBC, blocking pathways energy supply, results 56.61% reduction ATP. addition, inhibition ATP production regulation can downregulate expression heat shock proteins (HSPs) sensitized PTT, HSP70 level HSP90 are downregulated 25.68% 41.89%, respectively. This study provides new insights into combining PTT TNBC.

Breast cancer is the leading cause of cancer-related deaths in females worldwide, and liver one most common sites distant metastases breast patients. Patients with face limited treatment options, drug resistance highly prevalent, to a poor prognosis short survival. Liver respond extremely poorly immunotherapy have shown treatments such as chemotherapy targeted therapies. Therefore, develop optimize strategies well explore potential therapeutic approaches, it crucial understand mechanisms In this review, we summarize recent advances research discuss their for improving patient prognoses outcomes.

A key player in mitochondrial respiration, p32, often referred to as C1QBP, is mostly found the matrix. Previously, we showed that p32 interacts with DLAT mitochondria. Here, expression was reduced ccRCC and suppressed progression metastasis animal models. We observed increasing led an increase oxidative phosphorylation by interacting DLAT, thus, regulating activation of pyruvate dehydrogenase complex (PDHc). Mechanistically, expression, concert suppresses PDHc activity TCA cycle. Furthermore, our research discovered has a direct binding affinity for copper, facilitating copper-induced oligomerization lipo-DLAT specifically cells. This finding reveals innovative function p32/DLAT/copper glycometabolism cycle ccRCC. Importantly, provides important new understandings underlying molecular processes causing abnormal metabolism linked this cancer.
