Autoimmune rheumatic diseases (ARDs) are a heterogeneous group of conditions characterized by excessive and misdirected immune responses against the body's own musculoskeletal tissues. Their exact aetiology remains unclear, with genetic, demographic, behavioural environmental factors implicated in disease onset. One prominent hypothesis for initial breach tolerance (leading to autoimmunity) is molecular mimicry, which describes structural or sequence similarities between human microbial proteins (mimotopes). This similarity can lead cross-reactive antibodies T-cell receptors, resulting an response autoantigens. Both commensal microbes microbiome pathogens trigger thereby potentially contributing onset ARDs. In this review, we focus on role mimicry rheumatoid arthritis systemic lupus erythematosus. Moreover, implications also briefly discussed ankylosing spondylitis, sclerosis myositis.

Human longevity is a sex-biased process in which sex chromosomes and sex-specific immunity may play crucial role the health lifespan disparities between men women. Generally, women have higher life expectancy than men, exhibiting lower infection rates for broad range of pathogens, results prevalence female centenarians compared to males. Investigation immunological changes that occur during healthy aging, while taking into account differences sexes, can significantly enhance our understanding mechanisms underlie longevity. In this review, we aim summarize current knowledge on sexual dimorphism human immune system gut microbiome with particular focus centenarians, based exclusively data.

Introduction It has been reported that individuals with type 1 diabetes (T1D) are at a higher risk of concomitant occurrence other autoimmune diseases (AIDs). Currently, there is lack research investigating the causal relationship between T1D and AIDs. A comprehensive Mendelian randomization (MR) study was conducted using debiased inverse-variance weighted (dIVW) (IVW) estimators to examine bidirectional 12 Material methods Genome-wide association (GWAS) summary statistics datasets related or AIDs were obtained from FinnGen published cohort studies. Pruned SNPs in linkage disequilibrium (LD)-clumped single-nucleotide polymorphisms (SNPs) used as instrumental variables. For dIVW analysis, no genome-wide significance threshold applied for SNP selection. Results each 1-unit increase log-transformed odds ratio (OR) patients primary biliary cholangitis (PBC) rheumatoid arthritis (RA), ORs 1.123 (95% CI: 1.094–1.151) 1.133 1.100–1.167), respectively. Conversely, OR T1D, RA 1.383 1.213–-1.578). No associations found Conclusions Patients PBC have developing those also an increased RA. These findings highlight importance regular screening RA, PBC.

Abstract The current study was designed to explore the clinical significance of serum tissue inhibitor metalloproteinase 4 (TIMP4) levels in rheumatoid arthritis (RA). GSE1919 chip analyzed, differentially expressed genes (DEGs) were identified, and gene ontology as well Kyoto Encyclopedia Genes Genomes analyses identified DEGs conducted. Patients with RA ( n = 96) healthy individuals enrolled this study. Serum from participants collected, RT-qPCR WB have been conducted examine TIMP4 levels; additionally, interleukin (IL)-6 IL-1β determined using ELISA method. Abstract As global life expectancy increases, research reveals a critical challenge in aging: the progressive deterioration of immune function, termed immunosenescence. This age-related decline is characterized by complex dysregulation responses, which leaves older adults increasingly vulnerable to infections, chronic inflammatory states, and various degenerative diseases. Without intervention, immunosenescence significantly contributes morbidity mortality among elderly, intensifying healthcare burdens diminishing quality on both individual societal levels. review explores essential role zinc, trace element for health, mitigating impact slowing cascade immunological dysfunctions associated with aging. By modulating activity key cells pathways, zinc supplementation emerges as promising approach strengthen immunity, reduce oxidative stress, counteract "inflammaging," state chronic, low-grade inflammation that accelerates tissue damage drives disease progression. Zinc’s involvement cellular defense repair mechanisms across system highlights its ability enhance cell functionality, resilience, adaptability, strengthening body's resistance infection manage stressors contribute diseases Indeed, has demonstrated potential improve decrease inflammation, mitigate risk conditions including diabetes, depression, cardiovascular disease, vision loss. Given prevalent barriers adequate intake adults, dietary limitations, decreased absorption, interactions medications, this underscores urgent need address deficiency aging populations. Recent findings zinc’s molecular effects health present practical, accessible intervention supporting healthier improving life. integrating into targeted strategies, public efforts may not only sustain immunity elderly but also extend healthy longevity, costs, potentially incidence strain systems worldwide.

Aging alters the immune system, leading to immunosenescence characterized by impaired T cell functions. The balance between regulatory cells and type 17 helper (Th17) is crucial for maintaining peripheral homeostasis. disrupts this balance, contributing a systemic chronic proinflammatory environment that increases prevalence of age-related diseases. Treg/Th17 imbalance compromises self-tolerance, promoting autoimmune diseases such as rheumatoid arthritis, lupus erythematosus, multiple sclerosis. Furthermore, inflammation driven aberrant responses significant risk factor progression cardiovascular (CVD), including hypertension, atherosclerosis, myocardial infarction, myocarditis. Autoimmune disorders further exacerbate CVD, which remains cause mortality among patients with This review provides an in-depth analysis mechanisms driving during aging, highlighting its impact on homeostasis, autoimmunity, health. It explores how inflammaging dysfunction contribute emphasizing shared pathways therapeutic strategies restore mitigate inflammation. Understanding these highlights potential restoring tolerance reduce inflammation, thereby mitigating onset conditions.
