ONOO– is mainly produced in mitochondria, and dysfunctional damaged mitochondria are degraded lysosomes through autophagy, so it important to synthesize a single probe for dual detection of mitophagy. Unfortunately, mitochondria-immobilized fluorescent probes mitophagy have not yet been developed. Hence, we first reported piperazine-based red-emitting (PMR), which only can detect but also could be used image cellular by the pH variations because protonation piperazine moiety. PMR was designed prepared introducing ring as response group, lipophilic cation targeting moiety, benzyl chloride immobilizing mitochondrial proteins thiol groups. displayed an enhanced fluorescence at 640 nm acidification. Using these advantages PMR, successfully visualizing process induced rapamycin or starvation, chloroquine inhibit rapamycin-induced prevent fusion autophagosomes lysosomes. showed good sensitivity with limit 23 nM ONOO–, applied imaging exogenous/endogenous ONOO–. Combining above design, may study detailed function ONOO–-associated physiological pathological processes.
