Cytoarchitectural staining is of great importance in disease diagnosis and cell biology research. This study developed user-friendly multifunctional red-emissive carbon dots (R-CDs) for rapid nucleus via targeting nuclear proteins. R-CDs, simply prepared by electrochemical treatment 1,2,4-benzenetriamine, exhibit strong emission at 635 nm when excited 507 nm. The R-CDs can rapidly stain the human SH-SY5Y, HepG2, HUH-7 cells with a high signal-to-noise ratio owing to fluorescence enhancement after entering nucleus. Compared conventional cytosolic dyes such as Hoechst DAPI, are cheaper, more highly dispersed water, stable (requiring no stringent storage conditions). show optical properties insignificant photobleaching over 7 days salt resistance up 2 M NaCl. More importantly, possessing positive charge, allow live (3 min) dead (10 s) saline. According kinetic variation, distinguish from cells. Hypochlorous acid can be employed as a biomarker for blood infection (such sepsis) and tissue damage drug-induced liver injury, DILI), the diagnosis of or achieved through detection hypochlorous in relevant biological samples. Cancer is a serious global health threat, and photodynamic therapy (PDT) an effective treatment method for cancer. This works by generating large amount of singlet oxygen (1O2) under the influence light, which induces apoptosis in tumor cells, leading to their destruction. However, resistance cells limits development PDT, thus combination ferroptosis provides new perspective PDT. During PDT ferroptosis, levels 1O2 microenvironment (viscosity) within often change. To address this, this study developed novel fluorescent probe, NI–QM-OH, based on TICT–ICT effect, capable monitoring changes viscosity during The probe exhibits excellent selectivity, high sensitivity, low LOD (0.38 μM), has been successfully applied bioimaging HepG2, HeLa, MCF-7 as well zebrafish. Most importantly, NI–QM–OH also enables visualization diagnosis fatty liver disease (both alcoholic nonalcoholic).

Hypochlorous acid exists as HClO in acidic conditions and ClO– alkaline conditions, posing a significant challenge for differentiation due to their strong closely similar oxidative reaction activities. Addressing this challenge, our study presents an asymmetric donor–acceptor–donor′ (D–A–D′) molecular architecture the design of fluorescent probe (PMT NPs) that demonstrates exceptionally high specificity toward alongside optimized ratiometric response. The incorporation electron acceptor 2-(diphenylmethylene)malononitrile (A) modulates reducing ability phenothiazine recognition site, adjusting probe's oxidation potential intermediate level between ClO–. This adjustment directly dictates selectivity, enabling it respond exclusively HClO. By incorporating D′, response shifts intramolecular charge transfer (ICT) from original D–A D′–A, instead usual Dox–A presented previous works. controls blue shift fluorescence wavelength upon recognition, thereby improving accuracy signals vivo. PMT NPs precisely recognize environments was validated through live cell imaging vivo experiments using zebrafish mouse models, real-time monitoring surges. dual-pronged strategy, which combines interaction modulation with D–A–D′ architecture, promises revolutionize designs various biomolecules is anticipated advance understanding diseases linked these analytes.
