Pyroptosis, a specialized cell death mechanism crucial in cancer therapy, is distinct from apoptosis and primary necrosis. It linked to inflammation reactive oxygen species, particularly peroxynitrite (ONOO-), which implicated disease. Monitoring pyroptosis challenging due its connection with cellular polarity the tumor environment. In this research, we developed near-infrared probe, PBQI, highly sensitive responsive ONOO- fluctuations. PBQI targets mitochondria lipid droplets, enabling real-time tracking of pyroptosis-associated subcellular alterations. was found differentiate between normal cells during triggered by cisplatin metformin. To mitigate pyroptosis-induced inflammation, disulfiram used as an inhibitor, significantly alleviating excessive inflammation. This study highlights PBQI's utility studying interplay polarity, ONOO-, pyroptosis, contributes development more targeted therapies that optimize pyroptosis's antitumor benefits while minimizing inflammatory side effects.
