Deferasirox (DEF) is an important iron chelator for treatment overload related diseases. Monitoring DEF concentration in human serum will provide some valuable information clinical diagnosis and therapy. In this work, we developed a peroxidase-mimicking colorimetric sensor the detection of deferasirox by simple assembly telomeric dimeric G-Quadruplexes DNAzyme with Cu2+ ions. The catalyzed oxidation TMB presence H2O2 can generate quantitative signal, color change be discerned naked-eye. Compared monomeric G-Quadruplexes-Cu2+ DNAzyme, reaction rate significantly accelerated, gradually increased increasing number TTA spacer. Herein G2T3-Cu2+ chosen highly sensitive on basis DEF-Cu2+ complex-induced inhibition their activities. limit achieved down to 0.03 µM linear range from 0.05 1.2 µM. proposed strategy exhibits excellent selectivity potential interferent, such as metal ions small molecules. Importantly, robust enough perform well dispersible tablets samples. Overall, provides simple, low-cost rapid platform detection. This novel first example analysis utilizing signal amplification technology based G-Quadruplex which holds great quality control therapeutic drug monitoring.
