SUMMARY Excitatory/inhibitory (E/I) balance is thought to play a key role in cortical activity development. However, the modeling of networks with different E/I ratios not feasible vivo . To address this point, we modeled an vitro network deployed inhibitory neurons normally migrating from ventral telencephalon. Moreover, implemented striatal cultures and co-cultures mixed proportions neurons. The resulting contained various Parvalbumin (PV) + neurons, ranging 7% 73%. Interestingly, these pure cortical/striatal exhibited four distinct patterns spontaneous functional connectivity. Our findings highlighted critical for component developing correlated activity. Unexpectedly, PV were able generate appreciable burst due development strong inhibition, despite their lowest ratio. observations support notion that optimal ratio during essential establishment local capable generating spreading Highlights In neurogenesis models mouse devoid Cortical low neuron develops poor synchronized GABA inhibition unmask intrinsic ability highly Network response single node stimulus depends on A proper excitatory/inhibitory necessary Graphical Abstract
