Abstract Plants possess the ability to induce programmed cell death (PCD) in response abiotic and biotic stresses; nevertheless, evidence on PCD initiation during pear scald development involvement of trigger 6-methyl-5-hepten-2-one (MHO) this process is rudimentary. Pyrus bretschneideri Rehd. cv. ‘Dangshansuli’ was used validate such hypothesis. The results showed that superficial occurred after 120-d chilling exposure, which accompanied by typical PCD-associated morphological alterations, as plasmolysis, shrinkage, cytosolic nuclear condensation, vacuolar collapse, tonoplast disruption, subcellular organelle swelling, DNA fragmentation. These symptoms were aggravated MHO fumigation but alleviated diphenylamine (DPA) dipping. Through transcriptome assay, 24 out 146 PCD-related genes, transcribed cold storage, identified key candidate members responsible for these cellular biological alternations upon development. Among these, PbrCNGC1 , PbrGnai1 PbrACD6 PbrSOBIR1 implicated signaling pathway. Additionally, PbrWRKY2, 34 39 could bind W-box element promoter or activate their transcription, confirmed dual-luciferase, yeast one-hybrid, transient overexpression assays. Hence, our study confirms explores critical role process.

Clematis is the queen of vines, being an ornamental plant with high economic value. Waterlogging stress reduces value and limits its application. Melatonin plays important role in resistance to abiotic stresses. In this study, physiological responses gene expression levels two wild species, namely, tientaiensis lanuginosa , horticultural varieties, ‘Sen-No-Kaze’ ‘Viva Polonia,’ under waterlogging were analyzed determine effect melatonin on tolerance. The results showed that tolerances C. relatively poor, but significantly improved by concentrations 100 μmol·L -1 50 melatonin. Polonia’ had strong tolerance waterlogging, was 200 Under stress, relative conductivity H 2 O content increased significantly; photosynthetic parameters chlorophyll contents decreased; photosynthesis inhibited; soluble protein sugars decreased. Effective improvement after exogenous spraying, decreased 4.05%-27.44%; 3.84%-23.28%; 35.59%-103.36%; efficiency 25.42%-45.86%; antioxidant enzyme activities APX, POD, SOD, CAT 28.03%-158.61%; proline, protein, enhanced, cell homeostasis improved. Transcription sequencing performed differences tolerance, nine transcription factors selected highly correlated potential improve among which LBD4 MYB4 positively system, bHLH36 DOF36 WRKY4 negatively correlated. Photosynthetic capacity while MOF1 DOF47 REV1 ABR1 . could enhance flooding improving activity. This study provides basis reference for application waterlogging-resistant breeding

Apple scab, caused by the hemibiotrophic fungus Venturia inaequalis, is currently most common and damaging disease in apple orchards. Two strains of V. inaequalis (S755 Rs552) with different sensitivities to azole fungicides bacterial metabolite fengycin were compared determine mechanisms responsible for these differences. Antifungal activity tests showed that Rs552 had reduced sensitivity tebuconazole tetraconazole, as well alone or a binary mixture other lipopeptides (iturin A, pumilacidin, lichenysin). S755 was highly sensitive fengycin, whose close tebuconazole. Unlike from iturin family (mycosubtilin, A) similar on both strains, while those surfactin (lichenysin, pumilacidin) not active, except mixtures fengycin. The varies according their structure. Analyses difference azoles (which target CYP51 enzyme involved ergosterol biosynthesis pathway) linked (i) constitutive increased expression Cyp51A gene insertions upstream region (ii) greater efflux membrane pumps involvement ABC transporters. Microscopic observations revealed known interact plasma membranes, induced morphological cytological changes cells strains. Sterol phospholipid analyses higher level ergosta-7,22-dien-3-ol lower PI(C16:0/C18:1) S755. These differences could therefore influence composition explain differential However, antifungal activities mycosubtilin A two indirectly indicate sterols are probably resistance mechanism. This leads conclusion susceptibility studied.

In bilaterian animals, axon guidance decisions are regulated by many transmembrane receptor proteins called Roundabout (Robo) family members. During the developmental stages of fruit flies (Drosophila melanogaster), three Robo members play unique roles in central nervous system. Robo3 is revolutionarily conserved among taxa and studies show that regulates mediolateral axonal navigation. Recent suggest guides longitudinal axons a manner independent its ligand (slit). The expression patterns controlled transcription factors (TFs) significant role gene regulation, it not fully understood mechanism. Knowing factor binding sites (TFBS) would help to predict TFs regulate Robo3. this study, bioinformatics tools MEME Suite, TOMTOM, MAST were utilized analyze DNA sequence identify putative assist as docking regions for involved regulation expression. We found seven TFs: Btd, Opa, Mad, Odd, Twi, CF2, h. Mapping these TF motifs against showed located gene. Understanding implement novel strategies control overcome disorders related This study aims unknown may critical
