In inflammatory bowel diseases (IBDs), such as Crohn's disease (CD) and ulcerative colitis (UC), the immune system relentlessly attacks intestinal cells, causing recurrent tissue damage over lifetime of patients. The etiology IBD is complex multifactorial, involving environmental, microbiota, genetic, immunological factors that alter molecular basis organism. Among these, microbiota cells play pivotal roles; generates antigens recognized by antibodies, while autoantibodies target attack membrane, exacerbating inflammation damage. Given altered framework, analysis multiple biomarkers in patients proves exceedingly valuable for diagnosing prognosing IBD, including markers like C reactive protein fecal calprotectin. Upon detection classification patients, specific treatments are administered, ranging from conventional drugs to new biological therapies, antibodies neutralize molecules tumor necrosis factor (TNF) integrin. This review delves into targets, biomarkers, treatment options, monitoring techniques, and, ultimately, current challenges management.

Abstract One potential mechanism for microbiome-host, and microbiome constituents’ interaction communication involves extracellular vesicles (EVs). Here, the first time, we report capability of two M. smithii strains (ALI GRAZ-2), Candidatus intestini, Methanosphaera stadtmanae, as underrepresented components gut microbiome, to produce EVs. Interesting, size, morphology, composition AEVs were comparable bacterial EVs, indicated by ultrastructure, composition, proteomic metabolomic analyses; however, EVs substantially less prevalent in studied Archaea. When looking at proteomics more precisely, although from ALI intestini found be carrying unique proteins (n=135 n=30, respectively), shared within this genus (n=229), mostly adhesins(/like) proteins, or with IG-like domains. remarkable observation was uptake obtained stadtmanae Methanobrevibacter species human monocytes subsequent IL-8 secretion.

Abstract Many protein-protein interactions behave differently in biochemically purified forms as compared to their vivo states. As such, determining native protein structures may elucidate structural states previously unknown for even well-characterized proteins. Here we apply the bottom-up proteomics method, cryoID , toward a model methanogenic archaeon. While they are keystone organisms global carbon cycle and active members of human microbiome, there is general lack characterization methanogen enzyme structure function. Through approach, successfully reconstructed identified Methanosarcina acetivorans pyridoxal 5’-phosphate (PLP) synthase (PdxS) complex directly from cryogenic electron microscopy (cryoEM) images fractionated cellular lysate. We found that PdxS exists homo-dodecamer subunits, proposed supracomplex containing both glutaminase (PdxT) was not observed Our shows monomer fashions single 8α/8β TIM-barrel domain, surrounded by seven additional helices mediate solvent interface contacts. A density present at site cryoEM map interpreted ribose 5-phosphate. Abstract Microorganisms thriving in the human gastrointestinal microbiome have developed numerous mechanisms for microbiome-host and microbe-microbe interaction, including formation of extracellular vesicles (EVs). However, interactive members archaeome remained largely unexplored. Here, we report capability two Methanobrevibacter strains M. smithii, Cand. intestini Methanosphaera stadtmanae, to produce EVs. While size (~ 130 nm) morphology these archaeal EVs were comparable bacterial EVs, proteomic metabolomic analyses revealed unique traits. The proteins found both smithii (n = 229) predominantly included adhesin adhesin-like proteins, involved archaea-surface archaea-bacteria interaction. Additionally, contained glutamic acid choline glycerophosphate, which could be gut-brain signalling. Vesicles from all studied species efficiently taken up by monocytes induced IL-8 production epithelial HT-29 cells. identification their components provides novel insights into complex interactions between host, potentially offering new paths modulate vesicle-mediated impacts on host health.
