Globally, widespread tuberculosis is one of the acute problems healthcare. Drug-resistant forms require a personalized approach to treatment. Currently, rapid methods for detecting drug resistance Mycobacterium (MTB) some antituberculosis drugs are often used and involve optical, electrochemical, or PCR-based assays. Despite large number these assays, it necessary develop new tests (for drug-resistant MTB strains) that structurally simple do not specialized equipment. Colorimetric assays involving colloidal solution gold nanoparticles (AuNPs) have good potential development needed diagnostic tools. Here, conditions were found formation tandem duplexes between DNA probes targets, representing part gene gyrA, either wildtype containing single-nucleotide polymorphism associated with fluoroquinolone MTB. Adsorption on AuNPs allowed distinguish two targets owing nano-constructs different structures. Interaction was analyzed by optical spectroscopy, dynamic light scattering, transmission electron microscopy. A scheme proposed direct colorimetric detection fluoroquinolone-resistance-associated at 2 nM concentration in liquid system based shift AuNPs’ absorption maximum.

The world's leading infectious disease killer tuberculosis (TB) has >10 million new cases and ~1.5 mortalities yearly. Effective TB control management depends on accurate timely diagnosis to improve treatment, curb transmission reduce the burden medical system. Current clinical diagnostic methods for face shortcomings of limited accuracy sensitivity, time consumption high cost equipment reagents. Nanomaterials have markedly enhanced specificity speed detection in recent years, owing their distinctive physical chemical features. They offer several biomolecular binding sites, enabling simultaneous identification multiple biomarkers. Biosensors utilizing nanomaterials are often compact, user‑friendly well‑suited detecting location settings with resources. present review aimed advances that occurred during last five years application diagnostics, focusing capabilities, structures, working principles significance key nanomaterials. current addressed limitations challenges nanomaterials‑based along potential solutions.
