Asthma is a chronic airway inflammatory disease that affects millions globally. Although glucocorticoids are mainstay of asthma treatment, subset patients show resistance to these therapies, resulting in poor control and increased morbidity. The complex mechanisms underlying steroid-resistant (SRA) involve Th1 Th17 lymphocyte activity, neutrophil recruitment, NLRP3 inflammasome activation. Recent studies provided evidence innate lymphoid cells type 3 (ILC3s) might be potential therapeutic targets for non-eosinophilic (NEA) SRA. Like cells, ILC3s play crucial roles immune responses, inflammation, tissue homeostasis, contributing severity corticosteroid NEA. Biologics targeting ILC3-related pathways have shown promise managing Th2-low asthma, suggesting new avenues SRA treatment. Abstract In the last decade, messenger ribonucleic acid (mRNA)-based drugs have gained great interest in both immunotherapy and non-immunogenic applications. This surge can be largely attributed to demonstration of distinct advantages offered by various mRNA molecules, alongside rapid advancements nucleic delivery systems. It is noteworthy that immunogenicity presents a double-edged sword. context immunotherapy, extra supplementation adjuvant generally required for induction robust immune responses. Conversely, non-immunotherapeutic scenarios, activation unwanted considering host tolerability high expression demand mRNA-encoded functional proteins. Herein, mainly focused on linear non-replicating mRNA, we overview preclinical clinical progress prospects medicines encompassing vaccines other therapeutics. We also highlight importance focusing host-specific variations, including age, gender, pathological condition, concurrent medication individual patient, maximized efficacy safety upon administration. Pseudouridine (Ψ) is an abundant RNA chemical modification that plays critical biological functions. Current Ψ detection methods are limited in identifying Ψs at base-resolution U-rich sequence contexts, where occurs frequently. Here we report "Mut-Ψ-seq" utilizes the classic N-cyclohexyl N′-(2-morpholinoethyl)carbodiimide (CMC) agent and evolved reverse transcriptase ("RT-1306") for mapping base-resolution. CMC selectively labels forming CMC-Ψ adduct show RT-1306 presents promoted read-through mutation against CMC-Ψ. We a high-confidence list of sites polyA-enriched RNAs from HEK-293T cells identified by orthogonal treatments (CMC bisulfite). The signatures resolve position UU-containing sequences, revealing diverse occurrence such sequences. Small nucleolar RNAs (snoRNAs) are one of the most abundant and evolutionary ancient group functional non-coding RNAs. They were originally described as guides post-transcriptional rRNA modifications, but emerging evidence suggests that snoRNAs fulfil an impressive variety cellular functions. To reveal true complexity snoRNA-dependent functions, we need to catalogue first complete repertoire in a given context. While systematic mapping characterization "snoRNAomes" for some species have been recently, this has not done hitherto zebrafish (Danio rerio). Using size-fractionated RNA sequencing data from adult tissues, created interactive "snoRNAome" database species.
