Imbalanced immune homeostasis in cancer microenvironment is a hallmark of cancer. Increasing evidence demonstrated that long non-coding RNAs (lncRNAs) have emerged as key regulatory molecules directly blocking the immunity cycle, apart from activating negative pathways for restraining tumor immunity. lncRNAs reshape via recruitment and activation innate adaptive lymphoid cells. In this review, we summarized versatile mechanisms implicated including inhibition antitumor T cell activation, blockade effector recruitment, disruption homing, immunosuppressive cells, inducing an imbalance between cells (cytotoxic lymphocytes, M1 macrophages, helper type 1 cells) versus (M2 2 myeloid derived suppressor infiltrate tumor. As such, would highlight potential novel targets immunotherapy.

Abstract Being a leading lethal malignancy worldwide, the pathophysiology of hepatocellular carcinoma (HCC) has gained lot interest. Yet, underlying mechanistic basis liver tumorigenesis is poorly understood. The role some coding genes and their respective translated proteins, then later on, noncoding RNAs (ncRNAs) such as microRNAs have been extensively studied in context HCC pathophysiology; however, implication long (lncRNAs) circular (circRNAs) indeed less investigated. Hepatocellular carcinoma (HCC) is a highly fatal malignancy with limited therapeutic options and high recurrence rates. Recently, immunotherapeutic agents such as immune checkpoint inhibitors (ICIs) have emerged new paradigm shift in oncology. Metastasis-associated lung adenocarcinoma transcript-1 (MALAT-1) is a long intergenic non-coding RNA (lncRNA) located on chr11q13. It overexpressed in several cancers and controls gene expression through chromatin modification, transcriptional regulation, post-transcriptional regulation. Importantly, MALAT-1 stimulates cell proliferation, migration, metastasis serves vital role driving the epithelial-to-mesenchymal transition (EMT), subsequently acquiring cancer stem cell-like properties developing drug resistance. modulates EMT by interacting with various intracellular signaling pathways, notably phosphoinositide 3-kinase (PI3K)/Akt Wnt/β-catenin pathways. also behaves like sponge for microRNAs, preventing their interaction target genes promoting EMT.
