Abstract Background Preprints are scientific articles that have not undergone the peer-review process. They allow latest evidence to be rapidly shared, however it is unclear whether they can confidently used for decision-making during a public health emergency. This study aimed compare data and quality of preprints released first four months 2022 mpox outbreak their published versions. Methods Eligible ( n = 76) posted between May August were identified through an established literature database followed July 2024 changes in publication status. Quality studies was assessed by two independent reviewers evaluate quality, using validated tools available design 33). Tools included Newcastle-Ottawa Scale; Assessment Diagnostic Accuracy Studies 2 (QUADAS-2); JBI Critical Appraisal Checklists. The questions each tool led overall assessment high (no concerns with design, conduct, and/or analysis), moderate (minor concerns) or low (several concerns). Changes (e.g. methods, outcomes, results) preprint-published pairs 60) one reviewer verified second. Results versions could evaluated 25 pairs) mostly as quality. Minimal no change from preprint identified: all observational (10/10), most case series (6/7) surveillance analyses (3/3) had while some diagnostic test accuracy (3/5) improved worsened scores. Among 60), outcomes often added version (58%) less commonly removed (18%). Numerical results changed 53% studies, these (22/32) minor did impact main conclusions study. Conclusions suggests minimal supports use preprints, same critical evaluation on applied

Monkeypox (Mpox) is a growing public health concern, with complex interactions within host systems contributing to its impact. This study employs multi-omics approaches uncover therapeutic targets and potential drug repurposing opportunities better understand Mpox's molecular pathogenesis. We developed an in silico host-pathogen interaction (HPI) network applied weighted gene co-expression analysis (WGCNA) explore between Mpox proteins. Subtype-specific protein-protein networks were constructed, key modules from the HPI WGCNA integrated identify significant To predict upstream signaling pathways transcription factors, we used eXpression2Kinases ChIP-X Enrichment Analysis. The multi-Steiner trees method was compare our findings those FDA-approved antiviral drugs. Analysis of 55 differentially expressed genes infection revealed 11 kinases 15 factors as regulators. identified 16 targets, categorized into 8 proviral (ESR2, ERK1, ERK2, P38, JNK1, CDK4, GSK3B, STAT3) designated for inhibition, (IKKA, HDAC1, HIPK2, TF65, CSK21, ESR2, GSK3B) activation. Proviral are involved AKT, Wnt, STAT3 pathways, while impact AP-1, NF-κB, apoptosis, IFN pathways. Promising candidates identified, including kinase inhibitors, steroid hormone receptor agonists, notably Niclosamide. enhances understanding by identifying repurposable drugs, providing valuable framework developing new treatments.

Mpox is a zoonotic infectious disease caused by the mpox virus (MPXV). Historically, majority of cases have been documented in Central Africa. However, since May 2022, there has notable rise reported from regions beyond Currently, over 110 countries spanning Europe, North America, South Asia, and other territories infections. This report details case involving patient who identifies as man sex with men (MSM) concurrently infected MPXV, human immunodeficiency type 1 (HIV-1), Pneumocystis jiroveci, well extensively drug-resistant tuberculosis (XDR-TB). had also received vaccination for smallpox past. Additionally, we provide photographic documentation charting progression dermatological manifestations associated mpox. highlights significance sexual intercourse crucial mode transmission The rapid widespread dissemination MPXV across various regions, especially among MSM communities, underscores importance enhancing preventive education efforts targeted at high-risk populations.

The vaccinia virus Tiantan (VTT) is widely utilized as a smallpox vaccine in China and holds significant importance the prevention of diseases stemming from poxvirus infections. Nevertheless, few studies have investigated influence VTT infection on host gene expression. In this study, we constructed time series transcriptomic profiles HeLa cells infected with both western reserve (WR) strains. We observed similar patterns viral expression, while expression levels genes varied between two There was an immediate repression particularly associated oxidative phosphorylation. Conversely, involved nerve growth factor (NGF)-stimulated transcription were significantly activated. upregulation linked to ribonucleic acid (RNA)-induced silencing complex (RISC) suggested potential role for posttranscriptional regulation interaction host. later stages infection, pathways such extracellular matrix organization, neutrophil degranulation, complement interferon responses, translation, programmed cell death are largely inhibited. A number exhibit correlations changes genes. that negatively correlated mainly enriched translation response infection. This study contributes advancing our understanding dynamics host, improving application VTTs facilitating development effective vaccines against monkeypox.
