In recent years, circular RNAs (circRNAs) have garnered significant attention due to their unique structure and function, positioning them as promising candidates for next-generation vaccines. The circRNA vaccine, an RNA offers advantages in preventing infectious diseases by serving a vector protein expression through non-canonical translation. Notably, vaccines demonstrated enduring antigenic generate larger percentage of neutralizing antibodies compared mRNA administered at the same dosage. Furthermore, can elicit robust cellular humoral immunity, indicating potential tumor vaccine development. However, certain challenges must be addressed facilitate widespread use both disease prevention treatment. These include low efficiency linear circularization, suboptimal targeting delivery systems, assessment side effects. This work aims describe characteristics functions circRNAs, elucidate mechanism behind vaccines, discuss applications treatment tumors, along with future applications.

Background Long non-coding RNAs (lncRNAs) in extracellular vesicles (EVs) have been confirmed as effective non-invasive biomarkers for multiple diseases. However, their expression and clinical value gastric cancer (GC) remain poorly understood. Materials methods Serum EV RNA was extracted from four patients with GC healthy controls, followed by high-throughput sequencing. LncRNAs were further validated training validation sets using quantitative real-time reverse transcription polymerase chain reaction. Results A total of 37,684 lncRNAs obtained, 10 selected based on the criteria ( P < 0.05 |log 2 FoldChange| ≥1). lncRNA RMRP, RPPH1, linc-ROR significantly higher than those chronic gastritis, atypical hyperplasia, or control (all 0.05). Three also correlated tumor diameter, lymphatic metastasis, distal TNM stage The area under curve (AUC) values 0.727, 0.774, 0.811, respectively. Corresponding sensitivity specificity 63.4% 85.4%, 50.7% 89.6%, 78.5% 66.7%. combination these three carcinoembryonic antigen (CEA) yielded an AUC 0.909, a 83.3% each. Furthermore, high RMRP levels associated worse disease-free survival overall (OS). Univariate multivariate Cox regression analyses that only independent prognostic factor GC. Finally, lncRNA-miRNA-mRNA network showed predicted to interact 15 miRNAs 69 mRNAs. In addition, immune cell infiltration, including neutrophils, central memory CD4 T cells, macrophage, natural kill cells. Conclusion are prospective biomarker infiltration It provides foundation development serum EV-targeted novel immunotherapy targets

Colon cancer (CC) is a common malignancy with rising incidence worldwide. Despite advances in treatment strategies, many patients still face poor prognosis due to the development of drug resistance. Long non-coding RNAs (lncRNAs) have emerged as important regulators various biological processes and been implicated progression. Among them, colorectal neoplasia differentially expressed (CRNDE) has drawn attention for its potential roles different cancers. However, specific functions CC remain unclear. In this study, we identified CRNDE highly CC, contributing tumor progression Mechanically, regulated by transcription factor TFAP2A. Additionally, inhibits pyroptosis, form programmed cell death, promoting ubiquitin-mediated degradation RIPK3, thereby reducing sensitivity cells 5-fluorouracil (5-FU). Our findings suggest that TFAP2A/CRNDE/RIPK3 axis plays critical colon chemoresistance, highlighting therapeutic targets improving outcomes.

Glioblastoma (GBM), one of the most aggressive brain tumors, has a 5-year survival rate less than 5%. Current standard therapies, including surgery, radiotherapy, and temozolomide (TMZ) chemotherapy, are limited by drug resistance blood-brain barrier. Integrating expression quantitative trait loci (eQTL) protein (pQTL) data shown promise in uncovering disease mechanisms therapeutic targets. This study combined eQTL pQTL analyses to identify potential GBM-related genes circulating plasma proteins for exploration. Using transcriptomic from The Cancer Genome Atlas (TCGA), we identified 2,528 differentially expressed genes, GPX7 CXCL10. eQTL-MR analysis identifies GBM-associated constructs protein-protein interaction (PPI) network.Integrating deCODE database, pQTL-MR, colocalization validated CXCL10.These findings provide new perspectives on GBM biology suggest actionable targets therapy. Despite limitations due sample size population-specific data, this highlights CXCL10 as promising candidates further investigation lays foundation targeted treatments.
