ABSTRACT Transcription activator‐like effectors (TALEs) mimic eukaryotic transcriptional activators and translocate into host plant cells via the bacterial type III secretion system (T3SS) during pathogenic interactions. They play a crucial role in disease development by regulating genes. Despite this, regulatory mechanisms which TALEs control OsWRKY transcription factors (TFs) remain poorly understood. In this study, we show that two from Xanthomonas oryzae pv. ( Xoo ) individually modulate TFs, resulting increased susceptibility reduced defense. Specifically, Xoo1219 Xoo2145 activate expression of OsWRKY104 OsWRKY55 , respectively, through direct increases to activating OsSWEET11 OsSWEET14 while suppresses defense against directly OsWRKY62 . These findings suggest hijack host's TFs create favorable environment for survival.

Abstract Bacterial Blight of Cotton (BBC) caused by Xanthomonas citri pv. malvacearum ( Xcm ) is an important and destructive disease affecting cotton plants. Transcription activator-like effectors (TALEs) released the pathogen regulate resistance to susceptibility. In this study, we sequenced whole genome Xss-V 2 -18 identified eight tal genes; seven on plasmids one chromosome. Deletion complementation experiments genes demonstrated that Tal1b required for full virulence cotton. Transcriptome profiling coupled with TALE-binding element prediction revealed targets GhSWEET15A04/D04 GhSWEET15D02 simultaneously. Expression analysis confirmed independent inducibility Tal1b, whereas additionally targeted Tal1. Moreover, GUS (β -glucuronidase Xa10 -mediated HR (hypersensitive response) assays indicated EBEs are direct specific activation candidate Tal1 Ta1b. These findings may advance our understanding dynamics between TALEs EBEs, decipher a simple effective DNA-binding mechanism could lead development more efficient methods gene editing transgenic research.
