Millions of people take hormonal contraceptives (HCs), often starting during adolescence when ovarian hormones influence brain and behavioral maturation. However, there is a fundamental lack information about the neurobehavioral consequences alterations via adolescent HC use. To begin addressing this gap, we validated rodent model administration characterized its impact on endocrine, transcriptional, endpoints. Cohorts intact post-pubertal female Sprague-Dawley rats received daily subcutaneous injections either vehicle or [10 μg ethinyl estradiol (EE) + 20 levonorgestrel (LNG)] for duration from postnatal day (PND) 35 to PND56. Blood tissue was collected at PND57. Other cohorts PND35 until assays were completed PND57-64. treatment effective, as vaginal lavage indicated disrupted estrous cycling ELISA suppressed serum luteinizing hormone in HC-treated rats. Liquid chromatography-mass spectrometry analysis showed EE LNG well diminished levels allopregnanolone testosterone NanoString nCounter that impacted expression genes related synapses, white matter, neuroimmune, monoamine, signaling hypothalamus medial prefrontal cortex. While no effects HCs seen sociability social preference test stress coping behavior forced swim test, risk-assessment behaviors novelty-induced hypophagia paradigm altered anxiety-like open field elevated plus maze. Overall, these data suggest exposure contraceptive critical developmental period may shape behavior.
