Abstract Analyses of the genetic diversity well-studied fungi Sordariales order, such as Neurospora spp. and Podospora anserina (syn. Triangularia ), have shown that species classically defined by morphology are often complexes cryptic species. Here, we report on delimitation among 76 strains producing mycelium sexual reproductive structures identical to those pseudo-homothallic Schizothecium tetrasporum Neoschizothecium ). Their whole genomes were sequenced well six closely related but eight-spored asci instead four-spored ones. The clustering based Average Nucleotide Identity (ANI) between identified eighteen grouped into three clades, which further supported a phylogenetic tree constructed with genome Single Polymorphisms (SNPs). Based their contrasting breeding systems large evolutionary distances, considered clades distinct complexes. Indeed, two them, pseudotetrasporum complexes, contains asci, while third one, named octosporum , heterothallic asci. Surprisingly it was nestled Our data reveals thus huge morpho-species convergent evolution pseudo-homothallism or reversion heterothallism within An epitype for sensus stricto is seventeen new formally described.

A bstract Just exactly which tree(s) should we assume when testing evolutionary hypotheses? This question has plagued comparative biologists for decades. Given a perfectly estimated tree (if this is even possible in practice), seldom know with certainty whether such truly best (or adequate) to represent the history of our studied traits. Regardless certainty, choosing required all phylogenetic methods. Yet, conflict and error are ubiquitous modern biology, still learning about their dangers hypotheses. Here investigated consequences gene tree-species mismatch regression presence incomplete lineage sorting. Our simulation experiments reveal excessively high false positive rates mismatched both small large trees, simple complex traits, known phylogenies. In some cases, find evidence directionality error: incorrectly assuming species traits that evolved according sometimes fares worse than opposite. To explore difficult yet realistic scenarios, also used rather trees conduct case studies, as well an expansive expression dataset investigate arguably best-case scenario one may have better chance match trait. Though never meant be panacea ail methods, found promise application robust estimator potential, albeit imperfect, solution issues raised by mismatch, perhaps offering path forward. Collectively, results emphasize importance careful study design highlighting need fully appreciate role adequate modeling

Abstract Thermophily is a trait scattered across the fungal tree of life, with its highest prevalence within three families (Chaetomiaceae, Thermoascaceae, and Trichocomaceae), as well some members phylum Mucoromycota. We examined 37 thermophilic thermotolerant species 42 mesophilic for this study identified thermophily ancestral state all prominent fungi. Thermophilic genomes were found to encode various thermostable enzymes, including carbohydrate-active enzymes such endoxylanases, which are useful many industrial applications. At same time, overall gene counts, especially in responsible microbial defense secondary metabolism, reduced thermophiles compared mesophiles. also reduction core genome size both Chaetomiaceae family Eurotiomycetes class. The Gene Ontology terms lost fungi include primary transporters, UV response, O-methyltransferases. Comparative genomics analysis revealed higher GC content third base codons (GC3) lower effective number than Furthermore, using Support Vector Machine classifier, we several Pfam domains capable discriminating between mesophiles 94% accuracy. Using AlphaFold2 predict protein structures endoxylanases (GH10), built similarity network based on structures. that disulfide bonds appears important structure, clusters correlate optimal activity temperature. Thus, comparative offers new insights into biology, adaptation, evolutionary history while providing parts list bioengineering

Biofilms represent a key challenge in the treatment of microbial infections; for instance, Staphylococcus aureus causes chronic or fatal infections by forming biofilms on medical devices. Herein, fungus Arcopilus navicularis was found to produce novel family PKS-NRPS metabolites that are able disrupt preformed S. aureus. Arcopilins A–F (1–6), tetramic acids, and arcopilin G (7), 2-pyridone, were elucidated using HR-ESI-MS one-dimensional (1D) two-dimensional (2D) nuclear magnetic resonance (NMR) spectroscopy. Their absolute configuration established synthesis MPTA-esters 2, analysis 1H–1H coupling constants, ROESY correlations, along with comparison crystal structure 7. Arcopilin A (1) not only effectively disrupts but also potentiates activity gentamicin vancomycin up 115- 31-fold times, respectively. Our findings demonstrate potential application arcopilins conjugated caused antibiotics unable biofilms.
