Abstract Background Heterorhabditis are important biological control agents in agriculture. Two populations, S8 and S10, were isolated from agricultural soils the United States of America. Molecular analyses, based on mitochondrial nuclear genes, showed that these populations conspecific represent a novel species “ Bacteriophora ” clade. This was named americana n. sp. is described this study. Methods To describe H. sp., we carried out phylogenetic reconstructions using multiple characterized their morphology, conducted self-crossing cross-hybridization experiments, identified symbiotic bacteria. Results molecularly morphologically similar to georgiana . Morphological differences between males include variations excretory pore position, gubernaculum size, gubernaculum-to-spicule length ratio, tail length, body diameter. Infective juveniles (IJs) differ IJs because have an invisible bacterial cell pouch posterior cardia small phasmid, whereas visible inconspicuous phasmid. Hermaphrodites differentiated by nerve ring distance anterior end, anal diameter, c′ ratio. Females can be females diameter Reproductive isolation confirmed, as does not produce viable offspring with any associated bacterium Photorhabdus kleinii Conclusions Based observed morphological morphometric differences, distinct placement, reproductive isolation, nematode isolates S10 species, which study provides detailed characterization contributing enhancing our knowledge diversity evolutionary relationships genus.
