The convergent evolution of coenzyme metabolism in methanogens provides critical insights into primitive life and metabolic adaptations. This study investigated the molecular functional dynamics eight coenzymes cofactors Methanosarcina mazei, a model methanogen essential for methane production energy conservation anaerobic environments. Phylogenetic genetic diversity analyses 706 protein sequences revealed conserved evolutionary trajectories interspersed with lineage-specific adaptations driven by gene duplication, horizontal transfer, selective pressures. Key findings included purifying selection methanofuran (Tajima's D = -2.9589) A -2.8555), indicating functions. B biosynthesis pathway showed balanced 2.38602), reflecting its plasticity. linked F420 biosynthetic enzymes closely to horonobensis, while F430 highlighted prokaryotic specialization distinct from their eukaryotes. Coenzyme M genes have demonstrated unique connections species across domains, such as Methanothermobacter thermautotrophicus Gekko japonicus, emphasizing broad adaptive significance. These reveal how M. mazei optimized pathways thrive extreme environments, bridging ancient systems Last Universal Common Ancestor contemporary ecological
