Real-time
monitoring
of
chemotherapy-induced
senescence
(CIS)
in
cancer
remains
a
challenging
task
that
would
lead
to
new
insights
into
the
adaptive
mechanisms
therapy
and
provide
guidance
for
management.
Here,
we
designed
tailor-made
nanoprobe
capable
imaging
CIS
sequential
activation
self-amplified
manner
by
reversing
senescence-related
impaired
ferritinophagy.
It
contains
an
amphipathic
polymer
as
spatially
responsive
vehicle,
Fe2+-activable
dye
reporter,
autophagy
inducer
signal
amplifier.
Owing
metabolic
changes,
preferentially
enriches
senescent
cells,
leading
situ
fluorescence
switching
reporter
labile
Fe2+.
Meanwhile,
restores
ferritinophagy
promotes
autophagic
degradation
accumulated
ferritin,
facilitating
conversion
ferritin-bound
iron
Fe2+
amplified
cells
yet
keeping
inert
nonsenescent
cells.
Of
note,
accumulation
sustained
ferritin
occur
at
same
subcellular
location,
thus
minimizing
diffusion
process-induced
nonspecific
responses.
The
feasibility
this
strategy
is
successfully
demonstrated
both
living
animal
models.
This
work
offers
way
therapeutic
evaluation
basic
understanding

treatment.

Clear
