Real-time monitoring of chemotherapy-induced senescence (CIS) in cancer remains a challenging task that would lead to new insights into the adaptive mechanisms therapy and provide guidance for management. Here, we designed tailor-made nanoprobe capable imaging CIS sequential activation self-amplified manner by reversing senescence-related impaired ferritinophagy. It contains an amphipathic polymer as spatially responsive vehicle, Fe2+-activable dye reporter, autophagy inducer signal amplifier. Owing metabolic changes, preferentially enriches senescent cells, leading situ fluorescence switching reporter labile Fe2+. Meanwhile, restores ferritinophagy promotes autophagic degradation accumulated ferritin, facilitating conversion ferritin-bound iron Fe2+ amplified cells yet keeping inert nonsenescent cells. Of note, accumulation sustained ferritin occur at same subcellular location, thus minimizing diffusion process-induced nonspecific responses. The feasibility this strategy is successfully demonstrated both living animal models. This work offers way therapeutic evaluation basic understanding roles treatment.

Clear delineation of tumor margins is essential for accurate resection and decreased recurrence rate in the clinic. Fluorescence imaging emerging as a promising alternative to traditional visual inspection by surgeons intraoperative imaging. However, probes lack accuracy diagnosis, making it difficult depict boundaries accurately. Herein, we proposed an offensive defensive integration (ODI) strategy based on "attack systems (invasive peptidase) defense (reductive microenvironment)" multi-dimensional characteristics design activatable fluorescent precisely. Screened out from series ODI strategy-based probes, ANQ performed better than unilateral correlation distinguishing between cells normal minimizing false-positive signals living metabolic organs. β-Galactosidase (β-gal) is the gold standard marker of cellular senescence, which linked with various age-related diseases. Therefore, it essential to develop more excellent probes that can real-time monitor β-gal activity in senescence vivo. Fluorescent/photoacoustic (FL/PA) dual-modal imaging possesses sensitivity and spatial resolution. To our knowledge, there has been no tumor-targeted FL/PA probe image by monitoring we developed a (Gal-HCy-Biotin) for β-gal-activatable tumor senescence. Gal-HCy without biotin used as control probe. Gal-HCy-Biotin superior due higher kinetic parameter than vitro. Moreover, could help enter accumulate cells signal. Abstract Cellular senescence is a steady state of cell cycle arrest necessary to maintain homeostasis in organisms. However, senescent cells may cause neighboring healthy cells, inducing the onset several diseases, such as inflammation, neurological disorders, and atherosclerosis. Therefore, early detection cellular extremely important. β‐Galactosidase (β‐gal), critical marker senescence, can be monitored facilitate diagnosis aging‐related diseases. Furthermore, β‐gal mainly found lysosomes, which have pH value about 4.5–5.5. Here, we developed near‐infrared fluorescent probe (QMOH‐Gal) for tracking vitro vivo via β‐gal. In addition, displayed high sensitivity specificity with good fluorescence signal acidity range. Subsequently, this QMOH‐Gal was successfully employed differentiate between normal by monitoring not only realized zebrafish but also palbociclib‐induced breast tumor senescence. Senescence is an intricate physiological progression that can be instigated by a multiplicity of factors. Aberrant cellular senescence capable precipitating substantial array diseases. During chemotherapy, drugs typically tend to gradually accumulate in the liver, thereby inducing liver and leading successive deterioration its function. β-galactosidase (β-gal), serving as significant index exploration senescence, has attracted considerable attention. In this study, fluorescence/photoacoustic (FL/PA) biomodal probe (Gal-QCS) was developed based on hemicyanine fluorophore for imaging β-gal process drug-induced senescence. Gal-QCS demonstrates rapid responsiveness, high sensitivity, remarkable selectivity detecting aqueous solutions. After incubation with β-gal, fluorescence signal at 810 nm significantly increases, concurrently, photoacoustic 775 also exhibits increment. Upon induction cell camptothecin, expeditiously selectively image senescent cells. Moreover, after administering mice FL/PA signals livers were enhanced 10.53-fold 1.43-fold, respectively.
