Angiotensin I, II, and III (Ang III) aldosterone (Aldo) play an important role in primary aldosteronism (PA) screening according to the study of blood pressure regulation mechanism. However, Ang III, Aldo are present human plasma at low concentrations have different polarities, which make it rather challenging for current detection methods simultaneously detect four analytes complex samples. In this study, a new magnetic covalent organic framework (COF) was synthesized enrichment plasma, method simultaneous developed based on COF liquid chromatography-tandem mass spectrometry (LC-MS/MS). The experimental results demonstrated that adsorption kinetics material conformed pseudo-second-order model, mechanisms were π-π stacking, electrostatic, hydrogen bonding interactions. Under optimized conditions, established has satisfactory linear ranges I: 100–25,000 pg/mL, II: 2–500 III: 3–750 Aldo: 20–5000 pg/mL), limit (0.8–5 high recoveries (93.0–111.3%), multiple recycling, superior those reported studies. Meanwhile, testing 20 clinical samples indicated effective could be used as biomarkers PA screening, proved feasibility enriching targets real prepared experiment provided reference design Aldo, LC-MS/MS idea greatly promoted development disease diagnosis expected further research.
