The development of precise and efficient delivery systems is pivotal for advancing CRISPR/Cas9 gene-editing technologies, particularly therapeutic applications. Engineered metal-organic frameworks (MOFs) have emerged as a promising class inorganic nonviral vectors, offering unique advantages such tunable porosity, high cargo-loading capacity, biocompatibility. This review explores the design application MOF-based nanoplatforms tailored targeted components, aiming to enhance precision efficiency. By incorporating stimuli-responsive linkers bioactive ligands, these MOFs enable controlled release payloads at target site. Although the role of kynurenic acid (KYNA) is not yet fully understood, recent research has implicated this tryptophan (Trp) metabolite as a significant biomarker in neurodegenerative diseases. In study, we developed an immunosensor platform based on self-assembled polyelectrolyte multilayers (PEMs), employing enzyme-labeled immunoreagent competitive displacement format that requires only single wash step. This enables detection KYNA and Trp with limits (LOD) 9 pg/mL 1.2 ng/mL, respectively. Results validated by traditional ELISA methods indicated elevated levels increased KYNA/Trp ratio cerebrospinal fluid (CSF) Alzheimer's patients compared to controls, consistent previous findings.
