Circulating tumor cells (CTCs) serve as valuable biomarkers for early cancer diagnosis in low abundance the bloodstream. Therefore, development of a biosensor ultrasensitive detection CTCs is imperative. Herein, an enzymatic biofuel cell (EBFC)-based self-powered has been developed which recognition with stimuli-responsive barcode probe (SRBP) triggers opening circuit "lock" and further activates DNA dual-signal amplification, achieving CTCs. The SRBP fabricated based on hyaluronic acid (HA)-modified ZIF-8 framework, functionalized Trigger H2 at certain ratio connected magnetic beads via hybridization between aptamer (Apt) HepG2 cells. specific target CTCs, hepatocellular carcinoma line G2 (HepG2) cells, by Apt results release SRBP. Upon stimuli glutathione (GSH) under weakly acidic conditions (pH 5-6), disintegrated, leading to releasing Zn2+, accompanied liberation Trigger. As result, that is, DNAzyme-mediated cyclic cleavage substrate-linked SiO2 (Sub-SiO2) bioanode catalytic hairpin assembly (CHA) biocathode, activated. This achieves limit 3 cells/mL excellent specificity, exhibits substantial potential cancers.
