DNA nanoassembly-loaded CFA@SD core-shell nanoparticles and doxorubicin (Dox) were constructed based on the rolling circle amplification reaction (RCA), which is called CFA@RD NAs. CFA NPs was prepared by coating a gold nanoshell surface of Fe3O4 modified with citrate. Then, single-stranded (SD, partial sequence complementary to RCA) further through gold-sulfur bond synthesize NPs. An RCA assembly (RA) integrating two different functional DNAs (S2.2 aptamer strand support strand) as basic skeleton. Based principle base pairing Dox/G-C autonomic binding, Dox loaded onto RA carrier construct CFA@RA-Dox nanoassembly (CFA@RD NAs). In vitro cell experiments showed that dual targeting active magnetic passive targeting) significantly enhanced uptake NAs MCF-7 cells reduced toxic side effects normal cells.
