An alternative metric to account for particulate matter (PM) composition-based toxicity is the ability of PM-species generate reactive oxygen species (ROS) and deplete antioxidants, so-called oxidative potential (OP). Acellular OP assays are most used worldwide, mainly those based on ascorbic acid (AA) dithiothreitol (DTT) depletion; values calculated from AA/DTT concentration over time kinetic curves. Since a great variability in OP-DTT OP-AA can be found literature, understanding factors affecting rate AA DTT oxidation presence PM-bound will improve interpretation values. In this work, study driven by usually PM (transition metals naphthoquinone (NQ)) was carried out. particular, influence Cu(II), Fe(II), Fe(III), Mn(II), Mn(III), 1,4-NQ, type fluid assay (phosphate buffer (PB), phosphate saline (PBS) artificial lysosomal (ALF)) analysed discussed. The reaction orders with respect active compound, constants were also determined. results show among studied depending used; mostly higher PB0.05M, followed PBS1x ALF. Moreover, different concentration-responses OP-DTT/OP-AA obtained. These differences explained obtained each compound fluid.
