Screening environmental pollutants that are harmful to the cardiometabolic status and understanding their key toxic pathways crucial for effective clinical intervention. Based on exposure data of 46 chemicals in a nationally representative 13,286 people, logistic regression mixture modeling were used preliminarily identify with significant impacts 12 indicators disorders. A total 15 found be associated integrated latent class, among which four (perfluorononanoic acid [PFNA], perfluorooctanoic [PFOA], thiocyanate, thallium) also contributed significantly effect. We constructed adverse outcome (AOPs) nine toxicants both models individual each disorder. Notably, fluoroalkyl substances affect multiple aspects hyperlipidemia by activating PPARα. performed molecular docking vitro experiments verify supplement toxicological mechanism PFNA. Through binding PPARα, PFNA increased levels downstream molecules including CD36 (fatty transfer), ACSL1 activation), CPT1a (intracellular transfer β-oxidation) ultimately promoted accumulation triglycerides lipid droplets HepG2 cells. These markers, together events other metabolic phenotypes, may potential targets scientific research or treatment.
