Therapeutic oligonucleotides are a novel chemical modality with unique characteristics that offer significant therapeutic potential. With the latest surge in development, they paving way to market at rapid pace. To support that, scalable and economically sustainable oligonucleotide production is required. The upstream part exploits well-studied phosphoramidite chemistry, known from 1960s, using automated synthesis on solid support. On other hand, downstream processing, which consists of purification isolation steps, has stayed shadow more thoroughly studied synthesis, even though it often regarded as bottleneck production. Therefore, this review aims highlight most important traditional approaches for ensure product quality, yields, scale, cost-efficiency well newly emerging separation technologies could become relevant future.
