Enhanced expression of carboxylesterase (CarE) genes is an important mechanism insecticide resistance in pests. However, their roles multi-insecticide have rarely been reported. Herein, two CarE (PxαE6 and PxαE9) were identified; relative levels three multi-insecticide-resistant populations P. xylostella (HN, GD-2017 GD-2019) 2.69- to 15.32-fold higher than those the sensitive population, they considerably overexpressed at larval stage midgut 4th instar. PxαE6 PxαE9 knockdown increased susceptibility GD-2019 larvae phoxim or/and beta-cypermethrin. The recombinant expressed Escherichia coli exhibited high hydrolysis activity towards α-NA. GC–MS LC–MS/MS assays revealed that could metabolize beta-cypermethrin with efficiency determinations 51.6% 21.1%, respectively, while 12.0%.
