Inflammatory bowel disease (IBD) refers to a cluster of intractable gastrointestinal disorders with an undetermined etiology and lack effective therapeutic agents. Amygdalin (Amy) is glycoside extracted from the seeds apricot other Rosaceae plants it exhibits wide range pharmacological properties. Here, effects mechanisms Amy on colitis were examined via 16S rRNA sequencing, ELISA, transmission electron microscopy, Western blot, immunofluorescence. The results showed that administration remarkably attenuated signs (reduced body weight, increased activity index, shortened colon length) histopathological damage in dextran sodium sulfate (DSS)-challenged mice. Further studies revealed significantly diminished DSS-triggered gut barrier dysfunction by lowering pro-inflammatory mediator levels, inhibiting oxidative stress, reducing intestinal epithelial apoptosis ferroptosis. Notably, lowered TLR4 expression phosphorylation proteins related NF-κB MAPK pathways. Furthermore, modulated balance flora, including selective rise abundance S24-7 decline Allobaculum, Oscillospira, Bacteroides, Sutterella, Shigella.
