ABSTRACT Resveratrol alleviates liver fibrosis in mice by upregulating IL‐10 to reprogram the macrophage phenotype; however, mechanism remains be elucidated. Building on our previous work, this study, we aimed determine role of TLR2/MyD88/ERK and NF‐κB/NLRP3 inflammasome pathways mediating effects resveratrol polarization. We investigated expression cytokines these a mouse model resveratrol‐treated macrophages. The results showed that TLR2, MyD88, ERK, NF‐κB1 tissues was increased fourth week after treatment with but decreased fifth week. Similar were obtained for pathway. also both macrophages elevated 24 h reduced 36 resveratrol. Immunofluorescence nucleocytoplasmic separation assays inhibited translocation NF‐κB from cytoplasm nucleus macrophages, associated inhibition Silencing ERK. In conclusion, are involved effect polarization subsequent modulation fibrosis. acts as common cytokine coordinates crosstalk between two pathways. Our findings highlight potential members therapeutic targets toward

Liver fibrosis is a common pathological feature of chronic hepatic injury that currently lacks effective therapeutic interventions. Albiflorin (ALB), pinane-type monoterpene derived from Paeonia lactiflora Pall, has notable anti-inflammatory and hepatoprotective effects. However, the potential role ALB against liver largely unknown. In this study, we discovered significantly inhibited CCl4-induced in mice. This was evidenced by improvements kidney function indexes, indicators, histopathological findings. vitro studies also showed TGF-β1-induced LX-2 cell activation reduced expression α-SMA collagen I. Additionally, found mitigates inflammation ameliorates targeting CXCL12/CXCR4 axis, as confirmed using CXCR4 inhibitor AMD3100 CCl4-treated Notably, combining with metformin (MET) enhanced inhibition progression. These findings highlight exerts anti-liver effects underscoring its standalone treatment or an adjuvant therapy.

ABSTRACT The plateau hypoxic environment is a main habitat for human beings, which can result in dysbiosis of the intestinal flora. Non‐steroidal anti‐inflammatory drugs (NSAIDs) are widely used that cause damage with long‐term administration. Moreover, administration these may exacerbate damage. This study aimed to investigate therapeutic effect resveratrol (RSV) on injury induced by NSAIDs rats under hypoxia. Aspirin was as inducer induce rats. Rats were divided into seven groups: Ck (vehicle group), HCk (high‐altitude control PAsp (plain aspirin‐treated HAsp (High‐altitude RSVL (low‐dose resveratrol‐treated RSVM (medium‐dose and RSVH (high‐dose group). body weight recorded every 7 days during experiment. On last day experiment, jejunal tissues collected hematoxylin eosin staining (H&E), feces analysis flora metabolite analysis. results demonstrated RSV inhibited loss initiated aspirin environment. it markedly elevated expression levels interleukin (IL)‐10 superoxide dismutase (SOD) while substantially reducing TNF‐α, IL‐1β, myeloperoxidase (MPO). Furthermore, 16SrRNA gene sequence showed both altered composition structure rat gut microbiomes. Metabolomics metabolites aspirin‐induced rats, content 2‐hydroxy‐3‐ (4‐hydroxyphenyl) protonic acid 3‐ [(1‐carboxyvinyl) oxy] benzoate increasing coumaryl, 3‐amino‐4 hydroxybenzoate, L‐carnitine. Resveratrol alleviate NSAID (Aspirin)‐induced regulating metabolites, best group (50 mg/kg).

IBD, which includes Crohn’s and ulcerative colitis, is associated with gut microbiota dysbiosis. The dysbiotic environment results in an elevation of harmful a diminution advantageous microbiota, leading to IBD. Interestingly, plant-based dietary compounds consisting fibers polyphenols have demonstrated promise be safe successful IBD treatment, studies revealing that they can improve dysbiosis, increase anti-inflammatory cytokines, decrease pro-inflammatory lower oxidative stress, barrier function. Plant-based shown potential reduce by regulating signaling pathways such as TGF-β/Smad, TRL-4/NF-κB/MAPK, TLR2-NF-κB, autophagy, pyroptosis, glycolysis/gluconeogenesis amino acid metabolism, Nrf-2/HO-1, microbiota-macrophage-arginine bile metabolism. Chronic liver injuries often lead to hepatic fibrosis, a condition characterized by excessive extracellular matrix accumulation and abnormal connective tissue hyperplasia. Without effective treatment, fibrosis can progress cirrhosis or hepatocellular carcinoma. Current treatments, including transplantation, are limited donor shortages high costs. As such, there is an urgent need for therapeutic strategies. This review focuses on the potential of plant-based therapeutics, particularly polyphenols, phenolic acids, flavonoids, in treating fibrosis. These compounds have demonstrated anti-fibrotic activities through various signaling pathways, TGF-β/Smad, AMPK/mTOR, Wnt/β-catenin, NF-κB, PI3K/AKT/mTOR, hedgehog pathways. Additionally, this highlights advancements nanoparticulate drug delivery systems that enhance pharmacokinetics, bioavailability, efficacy these bioactive compounds. Methodologically, synthesizes findings from recent studies, providing comprehensive analysis mechanisms benefits treatments. One hundred twenty male lambs a similar initial weight (15.5 ± 0.14 kg) were randomly divided into four groups with thirty per treatment: (1) H group (basal diet without RES or HMB); (2) H-RES (1.5 g/day RES); (3) H-HMB (1250 mg/day (4) H-RES-HMB 1250 HMB). The experiment was conducted for 100 days, including pre-test period 10 days formal 90 days. results showed significantly increased concentrations glutathione peroxidase, superoxide dismutase, IgM (p < 0.05), while malondialdehyde levels decreased 0.05). glycolytic indices creatinine kinase (CK), malate dehydrogenase (MDH), succinate (SDH) compared others A histological analysis that hepatic plate tissue appeared normal multiple cells. transcriptomic expression genes associated calcium signaling pathway (MYLK2, CYSLTR2, ADCY1, HRH1, ATP2B2, NOS2, HRC, ITPR1, CAMK2B) NF-κB (BCL2 CARD14) upregulated. key differential metabolites (d-pyroglutamic acid, DL-serine, DL-threonine, fumarate, glyceric acid) enriched pathways D-amino metabolism, citrate cycle (TCA cycle), carbon metabolism. combined non-targeted metabolomic analyses co-enrichment (NOS2 GLUD1) (fumarate) arginine biosynthesis-regulated activity, whereas (ME1, SCD5, FABP2, RXRG, CPT1B) (Leukotriene b4) co-enriched PPAR affected immune response by regulating PI3K/AKT cGMP/PKG signaling.
