Natural products (NPs) have attracted increasing attention due to their structural diversity, abundance, and low environmental impact, making them valuable for novel fungicide development. In this study, a series of coumarin-7-N/O acethydrazide derivatives were synthesized by incorporating acetic acid moieties into the coumarin scaffold. The in vitro antifungal activities these compounds evaluated against six common plant pathogenic fungi. Compound B13 demonstrated significant effects, with EC50 values 3.24, 1.89, 1.70, 0.36, 4.27, 1.50 μg/mL Botrytis cinerea, Alternaria solani, Fusarium graminearum, Rhizoctonia Colletotrichum orbiculare alternata, respectively. The discovery of novel protoporphyrinogen IX oxidase (PPO) inhibitors has become a key focus in herbicide development. To explore new PPO inhibitors, series N-phenylphthalimide derivatives with ether moieties were designed and successfully synthesized. Among these, compound B18 (Ki = 10.3 nM) exhibited strong inhibitory effect on NtPPO, outperforming flumiclorac-pentyl 46.3 flumioxazin 52.0 nM). It is noted that compounds A3, B18, B19, B20 showed broad-spectrum herbicidal activity against the tested weeds at 75 g a.i./ha. In addition, results molecular simulation density functional theory (DFT) calculations indicated possessed not only robust hydrogen bond Arg98 NtPPO but also superior chemical reactivity, electrostatic field, polarity. Therefore, may be regarded as promising lead for development high-efficiency inhibitors.

In continuation of our search for herbicide lead compounds with novel structures and enhanced activities, a total thirty 5-acylbarbituric acid derivatives containing maleimide moieties were designed synthesized, their herbicidal activities evaluated in the greenhouse. The bioassay results showed that some newly synthesized target had good activity, which BT-IV-1 displayed an excellent inhibitory effect on Brassia campestris, Amaranthus retroflexus, blitum, Chenopodium album, Portulaca oleracea, Abutilon theophrasti, inhibition rate > 80% at dosage 37.5 g ha-1, it was determined to be safe Oryza sativa, Zea mays, Panicum miliaceum 75 ha-1. Studying molecular mechanism by phenotypic observation, membrane permeability evaluation, docking, vitro maize protoporphyrinogen oxidase (PPO) activity evaluation reveals is PPO inhibitor. present work indicates can serve as potential compound further development PPO-inhibiting herbicides.
